Anemone vitifolia(Ranunculaceae)is widely distributed in Hubei,Anhui and Shanxi provinces of China.The rhizome of Anemone vitifolia mainly contains phenols,lignans and saponin compounds,and exhibits antibacterial,anti-inflammatory,anti-tumor and anti-rheumatic arthritis activities.As a national medicine,Anemone vitifolia has been utilized in many minority areas of China.But so far,there have been no reports on the chemical and pharmacological activities of Anemone vitifolia from Enshi of Hubei provinces,China.In this program,the chemical composition and anti-tumor activity of Anemone vitifolia.collected from Enshi were carried out for the first time to provide theoretical basis for its further development and utilization.Column chromatographies including silica gel,Sephadex LH-20,ODS and preparative HPLC were used to separate the constituents of the 70% EtOH extract of Anemone vitifolia.The structures were elucidated on the basis of spectroscopic data and chemical derivatization method,and screening the antitumor activity of compounds by MTT.The study led to the separation and identification of twenty-nine compounds,including phenylpropanoids,coumarins,organic acids,lignins,alkaloids and saponins.They are palmitic acid(1)? cinnamic acid(2)? benzoic acid(3)?4-methoxysalicylaldehyde(4)?dimethyl phthalate(5)?3-(4'-formylphenoxy)-4-methoxybenzalde-hyde(6)?eugenin(7)?methylcaffeate(8)?oleanic acid(9)?caffeic acid(10)?ferulic acid(11)?trachelogenin(12)?arctigenin(13)?scoparone(14)?7-methoxycoumarin(15)?1,8-dihydroxyanthraquinone(16)?chlorogenic acid(17)?astragalin(18)?isoquercitrin(19)?tiliroside(20)?tracheloside(21)?arctiin(22)?salicin(23)?thymine deoxyribose(24)?1-?-D-psicofuranosyluraci(25)?methyl-?-D-glucop-yranoside(26)?dioscin(27)?arbutin(28)?tachioside(29)?Compounds 3,4,5,6,7,8,12,13,15,16,20,21,23,25,26,28,29 were isolated from Anemone for the first time;except for compound 22,all compounds were isolated from Anemone vitifolia for the first time.In addition,compounds 12,13,17,18,19,20,21,22,23,27 were screened for antitumor activity,and the results showed the inhibition rate of diosgenin(27),arctigenin(12),arctiin(22)and salicin(23)against cervical cancer cells HeLa were87.9%,10.51%,7.0% and 0.5% at the concentration of 100 ?M.
