Triterpenoid saponins have been reported as the main chemical constituents of the plants of genus Anemone(family Ranunculaceae). The pharmacological studies have showed that they exhibited anti-tumor, anti-inflammatory and anti-bacteria activities. Anemone hupehensis Lem. belonging to this genus is mainly distributed in the mountain region in Southwest of China, it has been reported that thirteen triterpenoid saponins have been separated from A. hupehensis collected from Sichuan and Anhui Provinces etc., but no study on their anti-tumor activity has been reported to date. There are abundant resources of the plant throughout Hanzhong, Shangnan and Ankang districts of Shaanxi province. However, no investigation has been reported on the chemical constituents of this plant distributing in Shaanxi Province. The phytochemical study of the rhizome of A. hupehensis collected from Ankang district and the study on the anti-tumor activity of the isolated constituents were carried out for the first time, in order to distinguish the difference of the constituents from this plant distributing in different areas, clarify the chemical constituents of A. hupehensis in Shaanxi Province, and provide theoretical and experimental basis for the development of the medicinal resource.Column chromatographies(including silica gel, Sephadex LH-20 and ODS) and glycoside, 4-hydroxy-2-methoxypheny-l-β-D-glucopyranoside(23); one aromatic ester, dibutylphthalate(27). All the compounds except compounds 9 and 12 were isolated from A. hupehensis for the first time. And among them, compounds 1, 2, 3, 6, 19, and 25 are six new compounds. It should be mentioned that neolignan glycosides and simple phenylpropanoid glycosides were firstly found from the genus Anemone.The preliminary cytotoxic assay showed that, only the monodesmosidic saponin 10 of four triterpenoid saponins exhibited cytotoxic activity with IC50 values of 3.67 and 2.98 μM against Hep-G2 cells and U-87 MG, respectively. the other three saponins had be founded no cytotoxic activity; in neolignan glycosides, compounds 3 and 24 exhibited cytotoxic activity against Hep-G2 tumor cells and cytotoxic activity of the edge against U-87 MG cell, compounds 19 and 26 exhibited cytotoxic activity of the edge against Hep-G2 cell, other two simple phenylpropanoid glycosides had be founded no cytotoxic activity.Innovations and significance:(1) The systematic studies on the rhizome of Anemone hupehensis distributing in Shaanxi Province were carried out for the first time, and this led to the isolation and identification of twenty-seven compounds, including eleven triterpenoid saponins, eleven neolignan glycosides, three simple phenylpropanoid glycosides, one phenolic glycosides and one aromatic ester.(2) Six phenylpropanoid glycosides, the stereoisomers of known compounds, were elucidated as new compounds. Besides them, there are also another ten compounds separated from the genus for the first time. All the compounds except compounds 9 and 12 were firstly isolated from A. hupehensis.(3) Saponins which were identified in the study have been found in genus Amenone repeatedly. And this provided a chemotaxonomic significance for A. hupehensis. All the saponin except f compounds 9 and 12 were isolated from the plant for the first time. And this indicated that a difference really exsited in A. hupehensis distributing in different aeras, and accumulated new research data for the further studies on the saponin constituents of the plant.(4) A series of neolignan glycosides and simple phenylpropanoid glycosides have been founded with their absolute configurations identified in genus Amenone for the first time. This provided new evidence for the chemical composition of genus Amenone, and further revealed the difference of chemical consitituents from the title plant distributing in different areas, which might possess significant chemotaxonomic value.(5) On the base of the in vitro anti-tumor experiments, the preliminary structure-activity relationship of neolignans and saponins was discussed.(6) Our studies have expanded the pharmacodynamic material basis of Amenone hupehensis, and established a homebase for the rational utilization of the plant resources in Shaanxi Province. The studies also accumulated research materials for the further studies on the active substances of the plant, supplied new evidences for the further investigation of genus Amenone, and provided valuable leads for pursuing new drugs.
