Cancer threatens to human health seriously.The study of molecular mechanisms of antitumor drugs is the important pharmaceutical research topic.Ganoderma is rare traditional Chinese medicinal materials.Its immunomodulatory,antitumor,anti-aging,anti-oxidation,regulated blood sugar blood fat,and so on,has been widely recognized at home and abroad.Our group based on former study,studied the antitumor molecular mechanism of proteoglycan SBT from Ganoderma tsugae.The Western Blot method was operated to detect protein expression level,RT-PCR was carried out to measure expression of mRNA,and immunofluorescence technique was used to investigate membrane receptor changes.The results were shown that Laminarin,the inhibitor of Dectin-1,competitively inhibit SBT combine with Dectin-1 and reduce TNF-a protein and mRNA expression in the THP-1 cell.And inhibited the degradation of NF-κB and p65 subunit into nuclear.And also decrease the TLR4 gene transcription and protein expression.It was suggested that proteoglycan SBT induce THP-1 cell increased expression of TNF-a has correlation with SBT recognition to Dectin-1.The downstream pathway can be activated through SBT integrating with Dectin-1 receptor.And then,nuclear transcription factors NF-κB signal transduction pathways can be opened,induce the TNF-a protein expression to increase.In addition,the increase of TLR4 expresson is related with SBT combined with Dectin-1.Decin-1 is a major receptor of proteonglycan SBT.but not only.
