Varicocele Infertility In The Pathogenesis And Mechanism Of Bushenhuoxue Side

Objective: Probing into the tradition Chinese midical pathologenesis of sterility with varicocele. To study the pathogenesis of sterility with varicocele, and reveal the mechanisms of Bu Shen Huo Xue Prescription for sterility with varicocele.Methods: S.D male rats was divided into seven groups: sham group, model group, Bu Shen Huo Xue Prescription low dose group, Bu Shen Huo Xue Prescription middle dose group, Bu Shen Huo Xue Prescription high dose group, Shao Fu Zhu Yu Tang group, Wu Zi Yan Zhong Wan Group. Experimental sterility with varicocele was created in S.D male rats by partial ligation of left renal vein. Observe the changes of testis weight, testis elasticity, the equality of semen, the fertiliy of each groups. Take the Flow Cytometry to measure the apoptosis rate of spermatogenic cells and its quantity of apoptosis. Use the mthod of immunohistochemistry to detect the expression of Bcl-2 gene,Fas/FasL gene in testis. Observe the changes of histology and ultrastructure under light microscope and electron microscope. Use the chemistry method to measure the equality of MDA, SOD, NOS. Take the radioimmunochemistry method to measure the equality of TXB2,6-keto-PGFia in plasma.Results: The testis weight, testis elasticity, the equality of semen of model group rats decreased significantly, lose mainly fertility. Bu Shen Huo Xue Prescription improved the equality of semen and fertility of model rats. ㏕he apoptosis rate of spermatogenic cells of model rats increased significantly, the quantity of spermatogenic cells decreaed greatly. Bu Shen Huo Xue Prescription can reduce the apoptosis rate of spermatogenic cells, increase the quantity of spermatogenic cells. The expression of Bcl-2 gene in spermatogenic cells decreased greatly, the expression of Fas/fasL gene in testis increased greatly in model rats. Bu Shen Huo Xue Prescription can increase the expression of Bcl-2 gene in spermatogenic cells, decrease the expression of Fas/fasL gene in testis. 0 Model rats have obvious anomaly in histology and ultrastructure in testes. Bu Shen Huo Xue Prescription can relieve the impairment level of testes. ㏕he quantity of MDA, NOS in testes of model rats increased significantly, the quantity of SOD in testes of model rats decreased significantly. Bu Shen Huo Xue Prescription can reduce the lever of MDA and NOS, improve the lever of SOD in testes of model rats. The lever of TXB2 in plasma increased significantly, the lever of 6-keto-PGF(a have no significant change, but the odds of TXB2/6-keto-PGFia increased significantly. Bu Shen Huo Xue Prescription can reduce the lever of TXB2 and the odds of TXB2/6-keto-PGF,a.Conclusion: The study has suggested that the asthenia of kidney and blood stasis is the fundmental pathogenesis of sterility with varicocele. By the means of the experimental study, we detected that the great increasement of apoptosis ofspermatogenic cells in the testes of animal mode, the production of ROS increased and the capability of removing the ROS. The results suggest that the great mount of apoptosis of spermatogenic cells in the testes may be the pathogenesis of sterility with varicocele. The study of pharmacology of Bu Shen Huo Xue Prescription suggested that it can improve the quality of semen and fertility. Bu Shen Huo Xue Prescription can adjust the expression of the gene with relation to apoptosis, then affect the rate of apoptosis. Bu Shen Huo Xue Prescription can ameliorate the impairment of testes, relieve the impairment of ROS to testes.
