Objective:The research is designed to change the contents of OPN in the chondrocytes, in order to investigate the effect of OPN on the expression of IL-6and IL-8of chondrocytes in patients with osteoarthritis of the knee.Methods:The research was performed on16specimens of cartilage obtained from osteoarthritic patients who underment total knee arthoplasty surgery. All the cartilage samples were treated with collagenase2, then culture and purified obtain to chondrocytes in vitro,take the first passaged cells into three groups:(1)the control group,(2)treated with100ng/ml OPN for48h,(3)treated with1μg/mlOPN for48h, We designed and validated the real-time quantitative polymerase chain reaction (Q-PCR) assays with SYBR Green dyes for simultaneous quantification of the expression of the IL-6mRNA and IL-8mRNA.. All statistics of results were analysed by SPSS19.0.Results:1. Compared to control-group, OPN (100ng/ml) up-regulated IL-6mRNA expression in OA chondrocytes, and the difference showed statistically significant.(p<0.05).2. Compared to control-group, OPN (1μg/ml) up-regulated IL-6mRNA expression in OA chondrocytes, and the difference showed statistically significant.(p<0.05).3. Compared to OPN (100ng/ml), OPN (1μg/ml) up-regulated IL-6mRNA expression in OA chondrocytes, and the difference showed statistically significant.(p<0.05).4. IL-6mRNA expression in OA chondrocytes statistically significant direct correlated with the concentration of OPN.(r=0.59, p<0.01).5. Compared to control-group, OPN (100ng/ml) up-regulated IL-8mRNA expression in OA chondrocytes, and the difference showed statistically significant.(p<0.05).6. Compared to control-group, OPN (1μg/ml) up-regulated IL-8mRNA expression in OA chondrocytes, and the difference showed statistically significant.(p<0.05).7. Compared to OPN (100ng/ml), OPN (1μg/ml) up-regulated IL-8mRNA expression in OA chondrocytes, and the difference showed statistically significant.(p<0.05).8. IL-8mRNA expression in OA chondrocytes statistically significant direct correlated with the concentration of OPN.(r=0.59, p<0.01).Conclusion:1. OPN can up-regulate IL-6mRNA expression in OA chondrocytes, the high expression of IL-6mRNA in OA chondrocytes may be a result of inceased OPN level of OA cartilages.2. OPN can up-regulate IL-8mRNA expression in OA chondrocytes, the high expression of IL-8mRNA in OA chondrocytes may be a result of inceased OPN level of OA cartilages.
