Objective: To investigate the effects of Toll like receptors4(TLR4)/Nuclear factor kappa B (NF-ΚB) signaling pathway on the osteopontinexpression in fibroblast-like synoviocytes of human with knee osteoarthritis invitro. Methods: The study was performed on10specimens of synoviumobtained from osteoarthritic patients. Then the fibroblast-like synoviocyteswere cultured in vitro by collagenase digestion.All the3-5passaged cells weretaken and divided into four groups: blank control group, lipopolysaccharide(LPS) group, LPS+T LR4antibody group, LPS+NF-κ B inhibitor (PDTC)group. The levels of osteopontin mRNA in four groups were evaluated byreverse transcription-polymerase chain reaction(RT-PCR). Results: Comparedwith the blank control group, the osteopontin mRNA expression inosteoarthritis fibroblast-like synoviocytes was significantly increased inlipopolysaccharide (LPS) group (P <0.05). When TLR4antibody was added,the osteopontin mRNA expression was significantly lower thanlipopolysaccharide group. The addition of NF-κB inhibitor (PDTC)significantly reduced the expression of OPN mRNA while compared withlipopolysaccharide group. Conclusions: TLR4can promote the expression of osteopontin in osteoarthritis fibroblast-like synoviocytes by NF-κB signalingpathway.
