Objective:To establish of the animai model of disused osteoporosis in rat, and to study the observation stage of disuse osteoporosis. To study the effect of Yishen Hugu decoction on the changes of the blood and urine biochemical levels, the bone structure, bone biomechanical properties and the expression of bone morphogenetic protein-2(BMP-2) in the animal model of disused osteoporosis in rat.Methods:1.78SD rats of age3months were randomly divided into sham operation group, model group and blank control group, the rats in the group of model group were sciatic neurotomy.1,2,4,6,8,12weeks after operation,the blood and urine biochemical levels were measured, the bone mineral density (BMD),the femora biomechanical test and the tibia were decalcified and then stained with HE and Masson stain.2.70SD rats of age3months were randomly divided into7groups:normal group, sham operation group, model group, high dose of Yishen Hugu decoction group, medium dose of Yishen Hugu decoction group, low dose of Yishen Hugu decoction group and drug control group (Drynaria fortunei),each group had10rats.The rats in the groups of model group, high,medium,low dose of Yishen Hugu decoction group and drug control group were sciatic neurotomy.Normal group was given regular food and water supply,the rats in high,medium,low dose of Yishen Hugu decoction group were gavaged by a concentration of52g/kg,26g/kg,16.25g/kg dose of Yishen Hugu decoction respectively,and the rats in D drug control group were gavaged by a concentration of0.25g/kg dose of Drynaria fortunei. The blood and urine biochemical levels were measured, the femora biomechanical test, the tibia were decalcified and then stained with HE and Masson stain and the expression of BMP-2.Results:1. The differences of BGP, BMD,the numbers of nodes and free ends, bone biomechanical properties of the model group during different stages were significant statically (P<0.01),the differences of those indexes between1week after operation and other stages, between2week after operation and other stages are significant statically (P<0.05).The differences of the model group and blank control group were significant statically (P<0.05).The differences between sham-operating group and blank control group were not significant statically (P>0.05). From4weeks after operation (weeks4,6,8,12), those indicators, the model group were higher or lower than the corresponding time of sham operation group (P<0.05).2. The differences of ALP, BGP, Strap, HOP/Cr, BMD and the numbers of nodes and free ends of model group and sham operation group were significant statically (P<0.01). After were gavaged by Yishen Hugu decoction, those indexes of high, medium, low dose of Yishen Hugu decoction group were obviously higher or lower than that of model group (P<0.01). After were gavaged by Yishen Hugu decoction.the analysis of picture shows the positive index of the expression of bone morphogenetic protein-2of high,medium,low dose of Yishen Hugu decoction group were obviously higher than that of model group (P<0.01).Conclusion:Using sciatic neurotomy could establish an animal model of disused osteoporosis in rats,and significant disuse osteoporosis in four weeks after modeling. The disused osteoporosis can be prevented effectively by the Yishen Hugu decoction in the index of blood and urine biochemical levels, the bone structure, bone biomechanical properties and the expression of BMP-2of disuse osteoporosis in rats.
