Study On Effect Of Chinese Medicine Chang Yuning Granule On Jak/stat Signaling Pathway In Rat Models With Ulcerative Colitis

Objective:To discuss the basic pathogenesis of ulcerative colitis and investigate the regulating effects on JAK/STAT siganal pathway of active ulcerative colitis in rats of Chang yu ning granula which treating active ulcerative colitis with the effects of clearing away heat and toxic materials and strengthening spleen to remove dampness in order to clarify the functional mechanism of Chang yu ning Granula.90 Wistar rats were randomly divided into normal control group, model group, high dose group,middle dose group, low dose group and SASP group of 15.Making rats models according to the syndrome of accumulation of damp-heat by composite method.and then treating them with Chang Yuning Granula.SASP was given 0.5g/kg per day in western medicine control group; 4g/ml,2g/ml,1g/ml drug concentration were given 10ml/kg per day in high dose group, middle dose group and low dose group.After 4 weeks, to observe the pathological changes in rat gastric mucosa as well as JAK2,STAT3 expression and IL-6 level.Result:The expression of IL-6 in model group was significantly increased compared with normal control group;The expression of IL-6 in high dose group,middle dose group, low dose group and western medicine group was reduced compared with the model group, and the differences among groups were statistically significant, middle dose group and western medicine group had better effect compared with low dose group and high dose group, and the differences between two groups weren't statistically significant. The expression of JAK2 in model group was significantly increased compared with normal control group;The expression of JAK2 in high dose group,middle dose group,low dose group and western medicine group were reduced compared with the model group, and the differences among groups were statistically significant, the middle dose group and western medicine group had better effect compared with low dose group and high dose group, and the differences between two groups weren't statistically significant. The expression of STAT3 in model group was significantly increased compared with normal control group;The expression of STAT3 in high dose group,middle dose group,low dose group and western medicine group were reduced compared with the model group, and the differences among groups were statistically significant, middle dose group had the best effect among other dose groups, and the differences was statistically significant.Conclusion:1.Replicate rats models sucssessfully according to the syndrome of accumulation of damp-heat by using composite method;2. It Is indicated by statistical analysis that high levels of IL-6 in serum and the quantitive expression of JAK2,STAT3 in colonic mucosa exist.3.Chang yu ning granula with the function of clearing away heat and toxic materials and strengthening spleen to remove dampness can treat active ulcerative colitis,And improve the pathological situation in colonic mucosa.4. Chang yu ning granula can regulate JAK/STAT signal pathway by means of reducing the level of IL-6 and the expression of JAK2,STAT3 which may be the main mechanism on treating active ulcerative colitis.
