| Objective:To study the effect of the Uygur medicine Kuijie′an to the expression of STAT3 in intestinal tissue of rat ulcerative colitis (UC) model and detect the effect mechanism of Uygur medicine Kuijie′an to UC. Methods:The rats were divided into four groups at randomly, which was individually physiological saline group, 3 variant Uygur medicine Kuijie′an treatment group included large dose group ,medium and little dose group. After preparation of the ulcerative colitis rat models were induced by“DNCB+Acetic acid”, the whole condition were observed in all groups after 20 days treatment. The expression of STAT3 in the rat’s intestine was detected by semi-quantatitive reverse transcription-polymerase chain raction (RT-PCR) and Western blot technique.Results:At the level of mRNA: the expression of STAT3 in the large dose group was down-regulated than that of the physiological saline group(P<0.05). At the protein level: the expression of STAT3 have no statistic signifinance to compared with the physiological saline group(P>0.05); the expression of P-STAT3 in the medium and little dose group were down-regulated than that of the large group(P<0.05);there is no statistic signifinance when the physiological saline group is compared to the large dose group ,medium and little dose group(P>0.05).Conclusion:Uygur medicine Kuijie′an has conspicuous treatment effect in ulcerative colitis rat. STAT3 may participate in the occurrence and development of ulcerative colitis and Uyghur’s medicine Kui Jiean may cure the Ulcerative Colitis in rat by deregulating the expression of STAT3; The regulation of STAT3 may be happened on transcription level. |
