With the coming of the aging of population in the world, some important scientific subjects which include exploring the mechanism of aging, delaying the occurrence of aging and improving the quality of life towards the aged have been generally paid attention to. Beginging with the theory of traditional Chinese medicine and emperimental study, the research explores the pathogenesis of aging, aiming at seeking the effective method which can delay aging. Erzhi Pill is the typical prescription for invigorating yin of the liver and kidney, which is used as the object of research. The experimental study on Erzhi Pill was researched deeply on on the action and mechanism of delaying aging on sub-acute rat induced by D-galactose.Objective: To research the function and mechanism of Erzhi Pill in aspect of delaying aging.Methods: Discuss the essence of aging based on the theories of traditional Chinese medicine and modern datum. Sub-acute rat of senility made by D-galactose as experimental model, Liuwei Dihuang Pill as comparative Chinese medicine and vitamin-E as comparative western medicine, this subject carries out pharmacological research of Erzhi Pill which is classified the groups of lower,midst and high dosages in order to observe the general condition of rats and change of brain tissue morphology. Through detecting blood sugar levels of rats, blood fat contents, hemorheology changes, anti-oxidation capability, ionic transport of cell membrane capability and expression of brain tissue protein, the subject verifies the function of delaying aging of Erzhi PilPs each dosage towards sub-acute rats of senility and approaches the part mechanism of action.Results: (1) The liver and kidney have more important relation with aging, and invigorating yin of the liver and kidney has the action of delaying aging. (2) Erzhi Pill has the functions of significantly degrading contents of CHO, TG and LCL-C in blood serum caused by D-galactose leading to aging of rats and increasing HDL-C. The function of midst dosage expresses overall and predominant. (3) The midst and high dosages of Erzhi Pill have better functions of anti- hyperviscous status of whole blood and plasm. (4) The capability of anti-oxidation of Erzhi Pill is relatively overall, in general, the dosages of lower and midst groups can improve T-AOC,CAT,GSH-PX and SOD activity, degrade MDA,NO and NOS contents. (5) Each group of Erzhi Pill has the functions of improving activity of Ca2+ -Mg2+ -ATPase and Na+-K+-ATPase of liver cell membrane of aging rats in different extent. (6) Each group of Erzhi Pill has the functions of promoting Bcl-2 and inhibiting Bax proteinic expression, the effect of lower dosage is the best. Each group has the function of inhibiting Caspase-3 proteinic expression, but their discrepancies of statistical are not obvious. (7) Considering to brain tissue of rats, each treated group of Erzhi Pill has the action of recovery in different extent, their distinctions are not obvious in light microscope. The midst dosage of Erzhi Pill can obviously improve corpuscular ultrastructure with approaching normal level obversed under electronic microscope.Conclusions: Aging of human beings is always based on yin deficiency of the liver and kidney, so its treatment is invigorating yin of the liver and kidney firstly. Erzhi Pill is the typical prescription for invigorating yin of the liver and kidney calmly, which has the function of delaying aging with multistrata, multiway and multitarget. Through improving blood fat and abnormal status of hemorheology, increasing anti-oxidation capability and keeping histiocyte normal morphology, Erzhi Pill implements the action of anti-aging. Erzhi Pill has the actions of removing redundant oxygen free radical, reinforcing Na+-K+-ATPase and Ca2+-Mg2+-ATPase activity of cell membrane and regulating Bcl-2, Bax and Caspase-3 proteinic expression of brain tissue, all of these factors influence each other, the mechanism of delaying aging of Erzhi Pill might be relevant to these factors. In aspect of improving each index of every group, the midst and lower dosages are better, which illustrates the relations of dosage and effect of functions in the finite extent. |
