| Objective:To observe the effect of Zuogui Pill on the anti-oxidation capability ofsubacute aging rats induced by D-Galactose,authenticate that ZuoguiPill,which is an archaic formula with the role of anti-aging,contributes topostpone insenecence by means of experiment.And approach the function'smechanisms preliminarily.Methods:Duplicate the aging rats model by means of subacute insenecenceinduced by D-galactose,take Liuweidihuang Pill as positive control,observethe rats' common states and the change of the hippocamp nerve cell inultramicro-morphology and detect the activity of defensive antioxidases(T-AOC,CAT,GSH-PX,SOD),the concentration of free radical (H2O2,theability of restraining superoxide anion radical and hydroxy radical),ATPase,P16,Bcl-2,Bax and Caspase-3.To do these,authenticate the function thatlow,middle and high dose of Zuogui pill accommodate the anti-oxidationcapability of the subacute aging rats induced by D-galactose and that ZuoguiPill postpones insenecence.Consequence:Afer duplicating the model,many patho-representations occur in theaging rats including the worse general states,increasing active oxygen freeradical,lipid peroxidation and apoptosis,reducing antioxygen capability andionic transport capability in the cell envelope,overexpression of P16,impairing hippocamp nerve cell architecture and depositive lipofuscin inultramicro-morphology.After the intervention of the three doses of ZuoguiPill,occur many better patho-representations in the model rats including thenot too bad general states,the enhancing T-AOC and the elevating capabilityof restraining superoxide anion radical and hydroxy radical in the blood,the increasing CAT and GSH-PX in the liver and SOD in the liver and brain,therestraining MDA in the liver and brain and H2O2 in the blood and liver,theraising Na+-K+-ATPase and Ca2+-Mg2+-ATPase in the cell envelope of liverand brain,the up-regulating Bcl-2 and the down-regulating P16,Bax andCaspase-3 proteinum expression,the keeping lower grade of nerve celldamage and the reducing depositive lipofuscin in the hippocamp CA1.But thethree doses of Zuogui Pill have roles in different degree on these indexes.Conclusion:Zuogui Pill can relieve the model rats' aging state,elevate theanti-oxidation capability,decend the ROS,reduce the damage fromthe free radical on the cell envelop,raise the ionic transport capabilityof cell envlope,down-regulate the expression of P16,restrain apoptosis andremain the morphous and function of cell.Demonstrate that Zuogui Pill canpostpone insenecence and the above may be the function mechanisms.But theaggression of ROS can induce the damage of the architecture and function inthe cell,the depression of stability in the cell envelop,apoptosis anddepositive lipofuscin,elevating the anti-oxidation capability anddecending the damage from the ROS may be the most importantfunction mechanisms.Zuogui Pill reflects the traditional Chinese medicinetherapeutical principle of invigorating the kidney and replenishing essenceand these experimental consequences demonstrate the therapeutical principlecan be used to postpone insenecence.Although the three doses of Zuogui Pillhave roles in different degree on these indexes,the middle and high dose ofZuogui Pill have an overall and stable effect of therapy in general and the lowdose of Zuogui Pill has an oppositely unstable one.Demonstrate in somedegree the anti-aging function of Zuogui Pill has positive correlation with thedose. |
