In order to study the mechanism of tiao qi an shen fa, we contrast different point groups of tiao qi an shen fa to treat the chronic mild unpredictable stressors rats by acupuncture, electric acupuncture and moxibustion, hoping to find the best treatment of depression by acupuncture and moxibustion.1 we used "baihui, taichong" and "tanzhong, neiguan" points by acupuncture, electric acupuncture and moxibustion respectively to treat the depressed rats and observe the rats behaviors, the results shows that: Compared with control group, the increased weight, the sucrose preference and activity of rats in open-field test in model group decreased significantly; The treatment groups can improve behavioral disorders of depressed rats, and they were not clear differences.2 we used redioimmunoassay to observe the effects of acupuncture , electric acupuncture and moxibustion of points "baihui, taichong" and "tanzhong, neiguan" on HPA axis in the animal model of chronic stressed depression, the results shows that: Compared with control group, the serum CORT and ACTH level in model group and saline group increased significantly; The treatment groups can decrease the serum CORT and ACTH level; The level of serum CORT and ACTH in electric acupuncture group is lower than that in acupuncture group, while acupuncture group is lower than moxibustion group, the level of serum CORT and ACTH in "baihui, taichong" group decreased significantly, compared with "tanzhong, neiguan" group, the treatment groups haven't significant differences.3 we used redioimmunoassay to observe the effects of acupuncture , electric acupuncture and moxibustion of points"baihui> taichong" and "tanzhong> neiguan" on serum interleukin levels in the animal model of chronic stressed depression, the results shows that: Compared with control group, the serum IL-^ IL^ and TNF a level in model group and saline group increased significantly; The treatment groups can decrease the serum IL-2^ IL-6> and TNF a level; The level of serum IL^ IL-6% and TNF a in moxibustion group is lower than that in electric acupuncture group, while electric acupuncture group is lower than acupuncture group, the level of serum IL-2n IL-6> and TNF a in "tanzhongN neiguan" group decreased significantly, compared with "baihui> taichong" group, the treatment groups haven't significant differences.4 we used redioimmunoassay to observe the effects of acupuncture > electric acupuncture and moxibustion of points "baihui>taichong" and "tanzhong^eiguan" on cAMP level in serum and different districts of brain in the animal model of chronic stressed depression, the results shows that:In serum, compared with control group, the cAMP level in model group and saline group decreased significantly; The level of cAMP in acupuncture and electric acupuncture on "baihui n taichong" point groups are higher than that in model group, while the two groups haven't distinct differences; The level of cAMP in acupuncture and electric acupuncture on "tanzhongN neiguan" point groups and the two moxibustion groups haven't distinct differences, compared with model group; The level of cAMP in acupuncture group is higher than that in electric acupuncture group, the level of cAMP in "baihui -> taichong" group increased significantly, compared with "tanzhong> neiguan" group.In cortex, compared with control group, the cAMP level in model group and saline group decreased significantly; The level of cAMP in acupuncture and electric acupuncture on "baihui >taichong" and "tanzhongN neiguan" point groups are higher than that in model group, while the four groups haven't distinct differences; The level of cAMP in moxibution on "baihuu taichong" and "tanzhong> neiguan" point groups are higher than that in model group, but they haven't distinct differences; The level of cAMP in acupuncture group is higher than that in electric acupuncture group, the level of cAMP in "baihui ? taichong" group increased significantly, compared with "tanzhong^ neiguan" group.In hippocampus, compared with control group, the cAMP level in model group and saline group decreased significantly; The level of cAMP in acupuncture and electric acupuncture on "baihui > taichong" and "tanzhongN neiguan" point groups are higher than that in model group, while the four groups haven't distinct differences; The two moxibustion groups haven't distinct differences, compared with model group; The level of cAMP in acupuncture group is higher than that in electric acupuncture group, the level of cAMP in "baihuk taichong" group is higher than the that in "tanzhong> neiguan" group.In hypothalamus, compared with control group, the cAMP level in model group and saline group decreased significantly; The level of cAMP in acupuncture and electric acupuncture on "baihuk taichong" point groups and the two moxibustion groups are higher than that in model group; The level of cAMP in acupuncture and electric acupuncture on "tanzhong ^ neiguan" point groups are higher than that in model group, but the groups haven't distinct differences. The level of cAMP in moxibustion group is higher than that in acupunture group, while acupuncture group is higher than electric acupuncture group. The level of cAMP in "baihui n taichong" group increased significantly, compared with "tanzhong> neiguan" group.The level of cAMP in amitriptyline group has no distinct difference with saline group in seruirK cortex> hippocampus and hypothalamus.5 we used immunohistochemistry and insituhybridization to observe the changes of BDNF level and BDNFmRNA expression, the results shows that: we can see BDNF like immunoreactivity and signals of BDNFmRNA insituhybridization in all the groups' cortex and hippocampus CAi district. Compared with control group, the BDNF like immunoreactivity and signals of BDNFmRNA insituhybridization in model group and saline group were significantly decreased; The treatment group can increase the number of the BDNF like immunoreactivity and signals of BDNFmRNA insituhybridization in the depressed animal; The number of the BDNF like immunoreactivity and signals of BDNFmRNA insituhybridization in electric acupuncture group is more than that in acupuncture group, while acupuncture group is more than moxibustion group; The number of the BDNF like immunoreactivity and signals of BDNFmRNA insituhybridization in "baihui> taichong" group is more than that in "tanzhong^ neiguan" group.In accordance with above comprehensive analysis, we may draw the conclusion:(1) Acupuncture and moxibustion can improve behavioral disorders of depressed rats, increase the activity of depressed rats in open-field tesU the sucrose preference and the weight.(2) Acupuncture and moxibustion treat depression with regulating HPA axis.(3) Acupuncture and moxibustion treat depression with reducing the elevatory serum interleukins to regulate HPA axis.
