The Experimental Study on Jianpi and Shugan Prescription Regulating Th cells Differentiation and Expression of Signaling protein CaNObjective: Underlying antigen and multiple comprehensive factor, the CD4+ T cells recognize the MHC-peptide and costimulatory molecules on APC, polarize into two groups of functional and phenotypic differences, and regulate immune response. The first group is Th1, its hallmark is IFN-a,mediated cell-immune, which are responsible for delayed type hypersensitivity responses. Second is Th2, secrets IL-4 and mounting allergic responses. The balance between the Th1/Th2 subsets determines the type of response, and ultimately, the outcome of that response for an individual organism to any given pathogen. But if the Th1/Th2 imbalance, that can mediate au2.1toimmune diseases such as multiple sclerosis and arthritis. The fate of differentiating Th cells controlled by multiple signaling proteins and transcription factor, regulation is dependent on the balance of MAPkinase, protein kinase C, and calcineurin signaling after TCR engagement. Methods:1. Wistar rats, force to take Sijunzitang and Chaihushugansanprescription, bid a day, the fourth day feed 1 hour later, get their serum and filtrate to except the bactoria, 56 a hour, -20 storage.2. Take the Wistar rat's spleen, and isolate with nylon's column to obtain the T cells and B cells , treated the B cells by Mitomycin C, add Ova and ConA to activate the T cells, RPMI1640 with 15% prescription serum, culture for 24hr or 48hr.3. Extract the mRNA and plasmic protein, measure the IFN-a, IL-4, CaN mRNA by RT-PCR and Western boltting check the expression of CaN.Results:1. The observation by RT-PCR technology showed that lL-4, CaN mRNA are elevated at the 0.1% Ova group, so about of CaN by Western blotting, the chaihushugansan serum and sijunzitang serum all candownregulate it significantly. But there are no significance between the two groups.2. The IFN-a mRNA observed by RT-PCR technology is elevated at the 10% Ova group, sijunzitang serum can downregulate it significantly, and at the 0.1%Ova group prove it , Chaihushugansan serum have not effect on it significantly.ConeIution:1. Sijunzitang can increase the immune function effectively through enhancing the weak body, and maybe usefully treat the Th1-mediated autoinmmune response diseases such as inflammatory bowel disease.2. Chaihushugansan can regulate the mechanism of qi, and maybe treat the Th2-mediated autoimmune diseases effectively such as asthma.3. Our data shows that have more referential value of the selection of harmacotherapy on Qi deficiency syndrome and excess syndrome.
