Chronic bronchitis (called CB for short) belongs to the disease scope of "cough", "asthma", "retention of phlegm" and "distension of the lung" in light of traditional Chinese medicine (TCM). In the recent years, with the aging of population and increasing degree of the air pollution, the incidence of chronic bronchitis is on the rise and affects seriously the people's working ability as well as living quality. Therefore it's of great significance to make deeper research on its pathological mechanism and develop the effective medicine with the integration of both Chinese and western medical theory.Based on the conventional knowledge and the modern research achievements of TCM on chronic bronchitis, we developed Ke-Chuan-Ning capsule with the function of tonifying qi, promoting blood circulation, resolving phlegm and stopping asthma. In order to make probe into its treatment action mechanism, we adopted CB model of SD rats with smoking method, observed its effect on MDA, SOD, CAT, GSH, GSH-Px and studied the relationship between chronic bronchitis and imbalance of oxidation-anti-oxidation as well as the treatment function mechanism of Ke-Chuan-Ning capsule; By observing its effect on ET-1, NO, TXB2 and 6-Keto-PGF, a, We studied its function on qi-passage diastole, anti-spasm and blood flow; Furthermore, by observing its effect on such inflammation cellular factors as TNF, IL-16 and IL-8, we studied its anti-inflammation action. At the same time.we also observed its effect on the pathological form of bronchus and lung tissue as well as the expression of cNOS & iNOS under the condition of chronic bronchitis by adopting advanced technology including HE staining and immunohisto-chemistry.10I. Chronic Bronchitis and Imbalance between Oxidation and Anti-Oxidation as well as the Treatment Function Mechanism of Ke-Chuan-Ning CapsuleWe randomly distributed 40 SD rats into model group of Chronic bronchitis (Model group for short), low-dose group of Ke-Chuan-Ning (Low-dose group for short), high-dose group of Ke-Chuan-Ning (High-dose group for short), control group of Guilong Ke-Chuan-Ning (Guilong group for short, a kind of positive control Ke-Chuan-Ning capsule) and normal group without any disposal (Normal group for short). Then we put 32 rats in each model group into a special-made smoking room of 1m3, lighting the smoke consisting of 50g of paring, sawdust and tobacco leaves respectively twice per day, SOminutes per time, for 30 days on end, while the rats of the normal group were fed in non-smoking environment. And then we filled normal saline or Ke-Chuan-Ning capsule for 21 days through the stomach from the second day of the completion of model-made. 1) Normal group: filling lOml/kg of normal saline, once a day; 2) Model group: filling lOml/kg of normal saline, once a day; 3) Low-dose group: filling 0.09g/ml of the suspension containing 0.9g/kg of the capsule once a day (14.1 times the dose of adult dose in clinic); 4) High-dose group: filling 0.18g/ml of the suspension containing 1.8g/kg of the capsule once a day (28.2 times the dose of adult dose in clinic); 5) Guilong group: filling 0.09g/ml of the suspension containing 0.9g/kg of the capsule once a day (14.1 times the dose of adult dose in clinic). At the second day after the three-week completion of stomach-filling, we made their blood plasma and blood serum, 10% of the lung tissue homogenate and BALF and stored them with the temperature of -20 癈 for examination.The result showed 1) Symptom observation of CB rats: the rats in each model group began to appear such symptoms as difficult respiration, wheeze, cough, lassitude, no luster of the hair, decreased food and water intake and much clear secretion flowing from the nasal cavity after the 12-day smoking; while the rats in normal group appeared no abnormal11reaction. 2) The effect of Ke-Chuan-Ning capsule on the MDA contents in the blood serum, lung tissue and BALF of CB rats: the MDA contents in the blood serum, lung tissue and BALF of CB rats in model group (3.63?0.30, 1.23 + 0.07, 4.
