| Objective:Psoriasis is a chronic inflammatory skin disease associated with multiple factors such as heredity and environment.Zhuhuang granules can improve psoriasis,but its related molecular mechanism is not very clear.Previous studies have found that phellodendron targeted genes are the most in the active ingredients of Zhuhuang granules.Therefore,the main purpose of this study is to explore the molecular mechanism of Phellodendri Chinensis Cortex,the main active ingredient of Zhuhuang Granules,in the treatment of psoriasis through animal experiments.Methods:Thirty mice were randomly divided into 5 groups:Control group was not treated.After the establishment of IMQ mouse model in the other groups,IMQ+HB Extract group,IMQ+HB Extract+Lv-shNC group and IMQ+HB Extract+Lv-shCTNNBIPl group were intragastrically administered with Cortex Phellodendri extract once a day for 7 days.The IMQ+HB Extract+Lv-shNC group and the IMQ+HB Extract+Lv-shCTNNBIPl group were subcutaneously injected with virus particles Lv-shNC and Lv-shCTNNBIP1 after the first gavage.The effects of Cortex Phellodendri extract and CTNNBIP1 knockdown on abnormal epidermal proliferation and secretion of inflammatory factors in mice were detected by PASI score,HE,IHC and ELISA.Results:1.Compared with the Control group,the PASI score of the IMQ group was significantly increased;compared with the IMQ group,the PASI score of the IMQ+HB Extract group was significantly reduced;compared with the IMQ+HB Extract+Lv-shNC group,the PASI score of the IMQ+HB Extract+Lv-shCTNNBIPl group was significantly increased.2.HE staining:The trend of skin thickness on the back of mice was the same as that of PASI score.3.IHC detection:Compared with the Control group,the protein level of CTNNBIP1 in the IMQ group was significantly decreased,and the protein levels of ki-67 and IL-17 A were significantly increased;compared with IMQ group,the protein level of CTNNBIP1 in IMQ+HB Extract group was significantly increased,and the protein levels of ki-67 and IL-17 A were significantly decreased.Compared with IMQ+HB Extract+Lv-shNC group,the protein level of CTNNBIP1 in IMQ+HB Extract+Lv-shCTNNBIPl group was significantly decreased,and the protein levels of ki-67 and IL-17A were significantly increased.4.ELISA detection:The trend of inflammatory factors in mouse tissues was the same as that of PASI score.The above results P<0.01,the difference was statistically significant.Conclusion:HB Extract inhibits Wnt Pathway and keratinocy te proliferation and inflammation by up-regulating CTNNBIP1,and ultimately improves psoriasis. |
