Cortex Phellodendri amurensis is the bark of phellodendron amurense Rupr. fromrutaceous plants. Alkaloid is the most active component and the most important alkaloidsare berberine and palmatine. In the2010edition of Chinese pharmacopoeia, the content ofberberine hydrochloride and palmatine hydrochloride are no less than0.60%and0.30%,respectively. There are two aspects of studies on cortex Phellodendri Amurensis in thepaper. On the one hand, study the technology on Cortex Phellodendri amurensis byultrahigh pressure extraction. On the other hand, study alkaloids contents from CortexPhellodendri amurensis by Quantiative Analysis of Multi-components by Single Marker(QAMS) and reference extract method.A new method using ultrahigh pressure was used to extract palmatine and berberinefrom the barks of cortex Phellodendri amurensis. The optimal conditions for ultrahighpressure extraction (UPE) of palmatine and berberine were quantifed by HPLC-DAD withstandard samples. Orthogonal design was applied to evaluate the effects of fourindependent factors (extraction pressure, extraction temperature, liquid/solid ratio andethanol concentration) on the yield. The optimal conditions of ultrahigh pressure extractionwere obtained at an extraction pressure of400MPa, extraction temperature of40℃,extraction time of4min, a liquid/solid ratio of30:1and an ethanol concentration of50%.The results showed that the UPE method can produce palmatine and berberine with thehighest yield. In addition, scanning electron microscope (SEM) images of the plant cellsafter ultrahigh pressure treatment were obtained to provide visual evidence of thedisruption ef ect. Compared with other extraction techniques, such as refux, ultrasonic, and soxhlet extraction, UPE shows several advantages, such as higher extraction yield, lessextraction time, lower extraction temperature and energy consumption.QAMS and reference extract method could solve the problems about lack andvaluableness of reference substance. And they are effective methods for assaying to buildquality standard of traditional Chinese medicine. The relative correction factors ofjatrorrhizine to berberine and palmatine to berberine were established and they were0.9868and0.9959, respectively. In this way, the calculated value was compared with theexternal standard method. There was no significant difference compared with the externalstandard method.Reference extracts were obtained by DAC in the experiment, and contained1.15%ofjatrorrhizine,13.94%of palmatine, and30.77%of berberine. Twenty kinds of CortexPhellodendri Amurensis were assayed by reference extract method and external standardmethod. There was no significant difference on two methods. Therefore, QAMS andreference extract method are viable and accurate for quality control when there is noreference substance.
