Purpose:(1)To investigate the changes of skin microbiome in psoriasis;(2)To explore the effect of Zhuhuang granules on skin microbiome of psoriasis.Methods:(1)18 mice were divided into control group,model group and Zhuhuang granule treatment group;(2)After a week of adaptive feeding,the mice in control group were gavaged with distilled water and coated with petroleum jelly;Mice in model group were intragastric with distilled water and coated with imiquimod cream;Mice in the treatment group were given Zhuhuang granules liquid intragastrically,and imiquimod cream was coated externally for consecutive 7 days.(3)On the 8th day,PASI score was performed on the three groups of mice,and pathological changes of skin tissue were observed by HE staining.The expression levels of IL-6,TFN-α and IL-1β in plasma and skin were detected by ELISA method,and the bacterial flora of mouse skin was detected and analyzed by 16 S r RNA sequencing technology.Results :(1)Compared with normal skin,the diversity and richness of bacterial flora in psoriatic skin were significantly decreased(P<0.01),and negatively correlated with PASI score,epidermal thickness,and the expression levels of inflammatory cytokine IL-6,TFN-α and IL-1β in plasma and skin.(2)There were obvious differences in the structure of the bacterial flora between the psoriatic skin and the normal skin.The psoriatic skin was mainly composed of staphylococcus species,and almost no other species could be detected.In addition to staphylococcus,the normal skin had a large proportion of bacteria such as copper bulide bacteria and staphylococcus tartaris.(3)After treatment with Zhuhuang granules,the diversity and richness of psoriasis skin microflora increased,the structure of skin microflora gradually recovered,the proportion of staphylococcus species decreased,and the proportion of copper glutinous bacteria increased,and the PASI score,epidermis thickness,and the expression levels of inflammatory factors IL-6,TFN-α,IL-1β in plasma and skin decreased.Conclusions:(1)Compared with the normal skin,the diversity and richness of the bacterial community in the psoriatic skin decreased and the structure changed.(2)Psoriasis skin flora may participate in the pathogenic process by changing the inflammatory state of blood or skin.(3)Zhuhuang granules had a benign regulation effect on the diversity,richness and structure of psoriasis skin flora.(4)Zhuhuang granules may improve the state of peripheral blood and skin inflammation by regulating skin microflora.
