Objective: The 16 S rRNA gene sequencing technology was used to analyze the differences of intestinal microflora among normal mice,imiquimod psoriasis model mice and Zhuhuang granule treated psoriasis model mice,and to explore the effect of Zhuhuang Granule on the changes of intestinal microflora in psoriasis mice.Methods: The mice were divided into imiquimod model group(D2group)and Zhuhuang granule treatment group(D3 group),and the control group(D1 group),with 6 mice in each group.Group D3 was given Zhuhuang granules by gavage,and groups D1 and D2 were given distilled water by gavage.After 7 days,the skin lesions,HE staining,PASI score,skin and serum inflammatory factors IL-1 ?,IL-6 and TNF-? levels of mice in each group were detected.At the same time,16 SrRNA gene sequencing analysis was detected in the feces of mice in each group,and the sequencing results were analyzed by cluster,diversity and abundance analysis.Results:(1)Comparison of skin lesions : compared with D2 group,the PASI score of D3 group was significantly decreased(P < 0.01).the pathological changes of D3 group such as hyperkeratosis,spinous layer hypertrophy and inflammatory infiltration were reduced.(2)Detection of inflammatory factors: the expression of IL-1 ?,IL-6 and TNF-? in skin and serum of D2 group was significantly higher than that of D1 group(P<0.01).The expression of IL-1 ?,IL-6 and TNF-? in skin and serum of D3 group was lower than that of D2 group(P< 0.0 1).(3)16S rRNA gene sequencing analysis: The differences of Observe,Chao1,ACE,Shannon,Simpson,J index among the three groups of intestinal flora were statistically significant.Lactobacillus ?Muribaculaceae S24-7 ? Alistipes in group D1 were significantly more than those in group D2,while the number of these three bacteria in treatment group D3 increased significantly after treatment of Zhuhuang granule.There was a linear relationship between PASI score,inflammatory factors and intestinal flora index.Conclusion:(1)Zhuhuang granule can reduce the inflammation of psoriasis mice induced by imiquimod.(2)There were differences in the total number,diversity,richness and evenness of intestinal flora among the 3 group.(3)Lactobacillus ?Muribaculaceae S24-7 ?Alistipes may play an inhibitory role in the pathogenesis of psoriasis,and Zhuhuang granule can achieve the therapeutic effect on psoriasis by increasing the content of these three bacteria.(4)There was a linear relationship between PASI score,inflammatory factors and intestinal flora index.
