Objective: Using data mining technology to systematically analyze the clinical medical records of Professor Zhang Shiqing in the treatment of Infantile Anorexia,and summarize Professor Zhang Shiqing’s medication rules and clinical experience in treating this disease.Then carry on the network pharmacology technology to the core drug group,so as to provide data support and famous TCM experience reference for the clinical treatment of infantile anorexia.Methods:(1)Collecting the IA medical records diagnosed and treated by Professor Zhang Shiqing in the affiliated Hospital of Gansu University of traditional Chinese Medicine from November 2020 to October 2022.Using Excel 2019,SPSS Modeler 18.0,SPSS Statistics26.0 and other software to perform frequency statistics,association rules,cluster analysis and factor analysis on medical record data that meet the standards;(2)For the core drug group obtained by data mining,the potential active ingredients and their action targets are obtained through the TCMSP,and the disease targets of anorexia in children are screened through the Gene Cards,TTD and OMIM.The protein-protein interaction(PPI)network model was constructed using the protein-protein interaction network database(STRING 11.5);Use Metascape platform to analyze GO function enrichment and KEGG pathway;Topology analysis and visualization are carried out by using Cytoscape 3.9.0 software.Results:(1)A total of 322 medical cases of IA were included in the data mining study.(1)Basic information of medical records: 187 patients were male(58.07%),135 patients were female(49.13%),the age ranges from 3 months to 14 years old,with a median age of 4 years,with the largest number of preschool children;(2)Prescription medication: Chinese medicines with high frequency of use include Hawthorn,Malt,Medicated Leaven,Membrane of Chicken Gizzard,baked Radix Glycyrrhizae,Radix Paeoniae Alba,etc;The association rule analysis showed that Mume Fructus-Radix Paeoniae Alba,Rhizoma Pinelliae Preparatum-Citri Reticulatae Pericarpium were commonly used drug pairs;High-frequency clustering analysis of traditional Chinese medicine can be divided into 8 categories,and factor analysis extracts 10 common factors with characteristic values greater than 1.0;The core drug groups were summarized as follows: Poria Cocos,Rhizoma Pinelliae,Citri Reticulatae Pericarpium,Mume Fructus and Radix Paeoniae.(2)A total of 39 potential active ingredients in the core drug group were screened in the network pharmacological research,including kaempferol,β-Sitosterol,carvedil,etc;231 potential action targets,with GAPDH,AKT1 and IL6 as core targets;GO enrichment analysis includes 2313 biological processes,151 cell components,260 molecular functions,and 211 pathways through KEGG analysis.Conclusion:(1)Data mining research: Professor Zhang Shiqing treats infantile anorexia with detailed differentiation of deficiency and reality,pays attention to the characteristics of children,uses both sweet and bitter drugs and mild and worm drugs,focuses on the spleen and stomach,attaches importance to the treatment from the liver,takes the method of invigorating the spleen as the principle,applies both medicine and supplement,and takes care of children’s stomach qi;The selection of prescription is based on the core drug group,which can be flexibly added or subtracted according to the certificate.(2)Network pharmacology research: It is speculated that the core drug group passes kaempferol β-The potential active ingredients,such as sitosterol and quercetin,regulate the core genes such as GAPDH,AKT1 and IL6,and then regulate PI3K-Akt,MAPK,calcium ion signal pathway,etc.,and jointly play a role in treating anorexia in children from multiple aspects,such as anti-inflammatory,improving gastrointestinal smooth muscle peristalsis,and promoting gastrointestinal hormone secretion. |
