Purpose:Using data mining technology and network pharmacology,studying the treatment of traditional Chinese medicine,mining the high frequency of traditional Chinese medicine,the combination of common medicine,and summarizing the medication rule of traditional Chinese medicine in the treatment of children,to explore a feasibility of traditional Chinese medicine in children.Data mining was used to analyze the medication rule of TCM treatment for tic disorder(TD),and to explore the action mechanism of high-frequency TCM treatment for TD based on network pharmacology.Material and method:1.In this study,we collected relevant literature on the treatment of Traditional Chinese medicine published from 2008 to January 2023,formulated literature inclusion and exclusion criteria,screened relevant literature,and established the "database of Traditional Chinese medicine for pediatric tic disorder".Data analysis was conducted based on the ancient and modern medical case cloud platform,and the data analysis methods such as drug statistics,drug clustering and association rules were used to mine high-frequency drugs,sexual taste return,prescription compatibility,etc.,and conduct visual analysis and display.2.The TCMSP database was used to search the chemical components of white peony root Chinese Chinese medicine,predict the protein target information,and establish the white peony root active ingredients-target database.Potential targets associated with tic disorder in children were searched in the Gene Cards,OMIM database.The targets were crossed between drug targets and disease targets,and the protein interaction network was constructed using the String platform.The network map of "TCM Active Ingredients-target" was constructed through Cyto Scape3.7.2,and DAVID was applied for GO functional enrichment analysis and KEGG enrichment analysis,and bubble maps were made using the microplatform.Results:1.Results of TCM treatment in children with tic disorders showed that the top 5high-frequency TCM were hook vine,white peony,licorice,silkworm and Gastrodia elata.The taste is sweet,bitter,xin,according to the liver,lung,spleen,heart.Multiple use of liver wind extinguishing drugs,tonic deficiency medicine,solution table drugs,heat clearing medicine,etc.Through the association rules,the higher support and confidence are the white peony root and Gastrodia elata.The drugs could be divided into 6 categories by cluster analysis,including 1 category: Zhuroru,Fructus aurantii,Pinellia pinellia,tangerine peel,poria cocos and atractylodes.2 classes: Gastrodia elata,Unguteng,cassia,scorpion;3categories: Bupleurum,Scutellaria scutellaria,rigidworm,cicada slough,chrysanthemum,white peony and licorice;4 categories: Ligusticum Chuanxiong,Parsnips and Angelica sinensis;Class 5: Acorus acorus,Radix polygonatum,turgid gold;Class 6: Keel,Oyster,Earthdragon,raw Rehmannia.2.Through TCMSP database and herb database,the high frequency drug 46 white peony root white peony root 33,13,7,3),181 targets,69 white peony root targets,383,93,2157 disease targets were collected and screened.The Wayn diagram shows 154 genes of white peony root and tic disorder.The five active components,including chitinase 、 Yohimbine 、p-hydroxybenzyl alcohol、suchilactone、Vanillyl alcohol,have analyzed the main core targets of protein interaction network,IL 1 B,ESR 1,CASP 3,EGFR,MYC,VE GF A,TNF,IL 6,and AKT 1.Disease-related pathways include MAPK signaling,the role of AGE-RAGE signaling in diabetic complications,Chagas disease,fluid shear stress and atherosclerosis,lipid and atherosclerosis,chemical carcinogenesis-receptor activation,Leishmania disease,human cytomegalovirus infection,EGFR tyrosine kinase inhibitor resistance,and IL-17 signaling pathway.Conclusion:1.Children tic disorder pathogenesis of traditional Chinese medicine movement is given priority to with liver wind,treatment from the liver,spleen,liver wind,spleen,phlegm for,commonly used treatment,complex,complicated,confidence and high support of medicine for hook-white peony root,hook-gastrodia elata,drug category and properties consistent with the basic pathogenesis of the disease,for children tic disorder clinical TCM syndrome differentiation treatment provide new methods and new ideas.2.In this study,the network pharmacology research method was used to preliminarily study the effective components,the target protein and the pathway of TD treatment in white peony root Gastrodia elata.The system reveals the active components of tic disorder Gene targets and signaling pathways,it is concluded that white peony root has a variety of components and targets,possibly through sedation,anticonvulsant and nerve protection... |
