| Objective: This study collected the electronic medical records of inpatients in the first affiliated Hospital of Guangxi University of traditional Chinese Medicine,collected the prescriptions of traditional Chinese medicine taken by patients with lumbar intervertebral disc protrusion to promote blood circulation and remove blood stasis,and conducted data mining on the composition of traditional Chinese medicine prescriptions.summarize the effective prescription rules,and analyze the core traditional Chinese medicine through the analysis of network pharmacology.To explore the potential targets and molecular mechanism of core traditional Chinese medicine components in the treatment of lumbar intervertebral disc protrusion,to provide reference for the clinical treatment of lumbar intervertebral disc protrusion,and to provide ideas and directions for follow-up experimental research and drug research and development.Methods: The electronic medical records of patients with "lumbar intervertebral disc protrusion" diagnosed as "lumbar intervertebral disc protrusion" and "low back pain-qi stagnation and blood stasis syndrome" diagnosed by traditional Chinese medicine in the first affiliated Hospital of Guangxi University of traditional Chinese Medicine were collected.The admission time was from January 2019 to December 2021.According to the inclusion criteria and exclusion criteria,patients were selected to take traditional Chinese medicine prescriptions with "promoting blood circulation and removing blood stasis" as the treatment principle.After data screening,cleaning and standardization,the data were collected and sorted into Excel software.The frequency,frequency and taste of traditional Chinese medicine were analyzed in Excel software,association rules analysis and cluster analysis were carried out by SPASS software,and the data were analyzed visually by cytoscape3.8.2 software.The above analysis results are summarized to get the core combination of traditional Chinese medicine.The core combination of traditional Chinese medicine obtained by data mining is analyzed by the method of network pharmacology,including the prediction of the main active components,core targets and related pathways of traditional Chinese medicine.First of all,the active components of traditional Chinese medicine are searched by the systematic Pharmacological Analysis platform of TCMSP,and then the targets of the effective components are predicted by Swiss Target Prediction database,and the gene names are standardized by Uniport.Secondly,by searching Gene Cards database,OMIM database and protein targets related to lumbar intervertebral disc protrusion.The two targets were compared and intersected,which is the potential target of core drug combination in the treatment of lumbar intervertebral disc protrusion.The visual network was made by Cytoscape3.8.2 software,and then the protein-protein interaction of the potential target was analyzed by String online analysis platform.Finally,the Metascape online analysis platform was used to analyze the GO biological function and KEGG metabolic pathway enrichment of the potential targets.Results: Collecting the prescriptions of traditional Chinese medicine for the treatment of lumbar intervertebral disc protrusion among inpatients,and through reading,comparison and data correction,a total of 330 inpatients were collected and 385 prescriptions of traditional Chinese medicine were recorded.The total frequency of traditional Chinese medicine involved is 5346.Traditional Chinese medicine is classified into 7 categories,including promoting blood circulation and tonifying drugs,and its effects are mainly promoting blood circulation and removing blood stasis and tonifying liver and kidney.The taste of traditional Chinese medicine is mainly bitter,sweet and pungent,the properties of traditional Chinese medicine are warm and mild,and the meridian tropism of traditional Chinese medicine is mainly liver meridian,kidney meridian and spleen meridian.A total of 21 groups of association rules are obtained through association rules.Six groups of cluster combinations were obtained by cluster analysis.Finally,the core combination of traditional Chinese medicine is Achyranthes bidentata and Eucommia ulmoides.Angelica sinensis,Erigeron Asarum.In the network pharmacology study,64 active components of the core traditional Chinese medicine combination were selected,including 689 active component targets and 545 disease targets related to lumbar intervertebral disc protrusion,of which 61 were related to the core traditional Chinese medicine.Through GO enrichment analysis,the enrichment results were 1355,including 1215 biological processes,43 cell components and 97 molecular functions.KEGG enrichment analysis showed that there were 162 pathways,including interleukin-17,tumor necrosis factor,MAPK signal pathway and so on.Conclusion:(1)This study uses data mining technology to study the prescription of traditional Chinese medicine for the treatment of prolapse of lumbar intervertebral disc by activating blood circulation and removing blood stasis in the first affiliated Hospital of Guangxi University of traditional Chinese Medicine in the past 3 years.Dig out the core combination of traditional Chinese medicine: Achyranthes bidentata,Eucommia ulmoides,Angelica sinensis,Erigeron breviscapus,the taste of traditional Chinese medicine is mainly bitter,sweet,pungent,warm and flat,and the meridians of liver,kidney and spleen are the main meridians.And the efficacy of most traditional Chinese medicine prescriptions is "promoting blood circulation and removing blood stasis,tonifying the liver and kidney",which is in line with the doctor’s rule of "invigorating the upright and dispelling evil,attacking and tonifying".It can provide reference for clinical TCM syndrome differentiation and treatment.(2)The core combination of traditional Chinese medicine summarized by network pharmacology data mining contains many effective components such as quercetin and kaempferol,and is related to IL-17、 MAPK、 NF-κ B、ECM、TLR4 and other signal pathways,which may have an impact on the process of lumbar disc degeneration and inflammation,which provides a reference for the clinical and experimental research of traditional Chinese medicine in the treatment of lumbar intervertebral disc protrusion. |
