Purpose:Data mining and network pharmacology research methods were used to explore the medication rules of traditional Chinese medicine in the treatment of diabetic cardiomyopathy(DCM).Mining high-frequency traditional Chinese medicines,commonly used drug pairs,drug combinations,etc.for the treatment of diabetic cardiomyopathy,and further through the method of network pharmacology,from different levels and perspectives of drug components,targets and pathways,to explore its mechanism of action in the treatment of diabetic cardiomyopathy.Provide theoretical basis for the treatment and follow-up research of the disease.Data and methods:1.In this study,the literatures related to the treatment of diabetic cardiomyopathy with traditional Chinese medicine were collected from databases such as CNKI,Wanfang,VIP,etc.from the establishment of the database to January 2022,and the inclusion and exclusion criteria were formulated,relevant literatures were screened,and a "TCM" was established.The Treatment of Diabetic Cardiomyopathy Database".Data analysis is carried out based on the ancient and modern medical record cloud platform,using the platform’s data analysis methods such as drug statistics,drug clustering,association rules,etc.,to mine high-frequency drugs,sex,taste,meridian,prescription compatibility,etc.,and visualize analysis and display.2.Use TCMSP database to retrieve the chemical constituents of Astragalus-Salvia miltiorrhiza Chinese medicine,predict the protein target information,and establish the active ingredient-target database of Astragalus-Salvia miltiorrhiza.Search Gene Cards,OMIM database for potential targets related to diabetic cardiomyopathy.Take the intersection of drug targets and disease targets,and use the String platform to build a protein interaction network.Cyto Scape3.7.2 was used to construct a network map of "active components of traditional Chinese medicine-targets",and DAVID was used for GO functional enrichment analysis and KEGG enrichment analysis,and the We Chat platform was used to make bubble charts.Results:1.According to the results of traditional Chinese medicine treatment of diabetic cardiomyopathy,the top 5 high-frequency traditional Chinese medicines are Astragalus,Salvia,Ophiopogon japonicus,Schisandra chinensis,and licorice.The nature and flavor are mainly warm,slightly cold,sweet,bitter,and acrid,and the heart,lung,and liver meridians are the main meridians.Use more blood-activating and stasis-removing medicines,qi-tonifying medicines,yin-tonifying medicines,heat-clearing medicines,etc.Through association rules,the ones with higher support and confidence are Astragalus-Salvia and Schisandra-Ophiopogon japonicus.Cluster analysis can be divided into 5 categories,1 category: Astragalus,Salvia;2 categories: ginseng,Ophiopogon japonicus,Schisandra;3 categories: Rehmannia glutinosa,Angelica,Chuanxiong,safflower,red peony,peach kernel;4 categories: licorice,Guizhi,Poria,Atractylodes;5 categories: Panax notoginseng,Pueraria,Pinellia,Cortex Moutan,Coptis;2.Through the TCMSP database screening,a total of 85 active components of Astragalus-Salvia miltiorrhiza(20 Astragalus and 65 Salvia miltiorrhiza)were obtained,461 corresponding targets of Astragalus and 932 targets of Salvia miltiorrhiza.1522 DCM disease targets.Through Venn diagram,it was concluded that there were 115 intersection genes of Astragalus-Dan involved in diabetic cardiomyopathy.Quercetin,luteolin,kaempferol,tanshinone IIA and other four active ingredients have many targets,and the main core targets of protein interaction network analysis are TNF,IL6,AKT1,VEGFA,IL1 B,TP53,CASP3,JUN,MMP9,PTGS2,etc.Disease-related pathways include cancer pathway,PI3K-Akt signaling pathway,proteoglycans in cancer,hepatitis B,tumor necrosis factor(TNF) signaling pathway,MAPK signaling pathway,HIF-1 signaling pathway,Fox O signaling pathway,and also There are NF-κB signaling pathway,insulin resistance,etc.Conclusion:1.The medication rule of traditional Chinese medicine in the treatment of diabetic cardiomyopathy,its medicinal properties are mainly warm,slightly cold,and flat,and the medicinal taste is mainly sweet,bitter,and acrid,and the meridians are related to the five internal organs.Drug classification is mainly based on blood-activating and stasis-removing drugs,qi-tonifying drugs,yin-tonifying drugs,and heat-clearing drugs.The drug pairs with high confidence and support are Astragalus-Salvia and Schisandra-Ophiopogon japonicus.The basic pathogenesis theory of the disease is consistent with the clinical symptoms of deficiency of both qi and yin,and blood stasis blocking collaterals.2.Using the research method of network pharmacology,systematically revealed the active components,gene targets and signaling pathways of Astragalus-Salvia in the treatment of diabetic cardiomyopathy,and concluded that Astragalus-Salvia can treat DCM through multiple components and multiple targets.It plays a role in improving myocardial apoptosis,anti-oxidative stress,inhibiting inflammatory response,and regulating glucose and lipid metabolism.
