Objective: To use data mining technology to explore the rules of prescription and drug use in the treatment of chronic obstructive pulmonary disease(COPD)with golden and water mutually reinforcing method,and use network pharmacology technology to explore the mechanism of core prescription in the treatment of COPD,in order to provide more theoretical basis and innovative ideas for the treatment or research of COPD.Materials and methods: With "chronic obstructive pulmonary disease AND pulmonary AND kidney " as the key word,the relevant documents were retrieved from CNKI,Vip and Wanfang databases,and the documents from June,1999 to December,2022.After screening the literature,Excel and SPSS were used to carry out frequency statistical analysis of drug use frequency,sex,taste and meridian tropism in the data.Then use IBM SPSS Modeler 18.1 and IBM SPSS Statistics25.0 software to carry out association analysis and cluster analysis on the drugs in the database to obtain the drug combination and core prescription.Collect the active ingredients and targets of the core prescription through the TCM database,and obtain the disease targets of COPD by querying the Dis Ge NET and Gen CLIP3 databases.The prediction target of "core prescription" and the prediction disease target of COPD are imported into the CTD platform to match the traditional Chinese medicine target and disease target,and the common target that the effective components of "core prescription" are closely related to COPD is obtained.Use Cytascape 3.7.2 software to build the network diagram of the effective active component-target of the "core prescription" treatment of COPD,conduct topological analysis on the network diagram,and obtain the important active components and main corresponding targets of the "core prescription" treatment of COPD.Input the key targets into STRING database,export the obtained data in TSV format and import it into Cytascape software,and finally get the protein-protein interaction network diagram.Finally,import key targets into DAVID database and select OFFICIAL_GENE_SYMBOL,select Gene List as the list type to conduct GO function enrichment analysis and KEGG signal pathway enrichment analysis.results:1.A total of 837 documents were retrieved.According to the inclusion criteria,a total of 204 prescriptions,181 traditional Chinese medicines and 2416 times of use were selected.Among them,there are20 drugs with a frequency of use greater than 32,while the top four are Astragalus(65.7%),Radix Rehmanniae Preparata(55.3%),Schisandra chinensis(48.6%),and Glycyrrhiza uralensis(48.0%).2.The total frequency of the four gases is 2434 times,ranking in the order of temperature,calm,cold,cool and hot.The frequency of temperature is 1254 times,51.9%,which is the most important drug property;The total frequency of the five flavors is 3508,and the rank of the five flavors is sweet,bitter,spicy,sour and salty.The frequency of sweet drugs was the first,1521 times,43.3%,followed by bitter drugs and bitter drugs,920 times and 732 times respectively;The total frequency of meridian tropism is 5402 times.The meridian tropism ranks in the following order: hand Taiyin lung meridian,foot Shaoyin kidney meridian,foot Taiyin spleen meridian,hand Shaoyin heart meridian,foot Jueyin liver meridian,foot Taiyang bladder meridian,and foot Yangming colon meridian.The drug frequency of hand Taiyin lung meridian was the highest,1352 times,25.0%,followed by foot Shaoyin kidney meridian,1021 times,17.5%,and the third was spleen meridian,947 times,17.5%.3.From the analysis of association rules,it can be seen that "astragalus membranaceus-orange peel" and "astragalus membranaceus-rehmannia chinensis-schisandra chinensis" are the most highly supported combinations of traditional Chinese medicine in the same group.Among the four association rules,the support of the first three combinations is relatively close,while the confidence of the combination of "astragalus membranaceus-rehmannia chinensis-licorice" is much higher,indicating that the combination is more closely related.From K-means clustering analysis,three new formulas can be obtained.One is astragalus,Radix Rehmanniae,Fructus Schisandrae,licorice,Poria cocos,Atractylodes macrocephala,Codonopsis pilosula,tangerine peel,Pinellia ternata,and Salvia miltiorrhiza;New formula 2:Astragalus,Radix Rehmanniae,Fructus Schisandrae,Poria cocos,Atractylodes macrocephala,Cornus officinalis,tangerine peel,Chinese yam;New prescription 3: Radix Astragali,Radix Rehmanniae Preparata,Fructus Schisandrae,Liquorice,Codonopsis pilosula.Through the analysis of association rules,cluster analysis and complex network analysis,the core Chinese medicine compound "Huangqi-Rehmannia-Schisandra chinensis" can be obtained.4.Using network pharmacology,it can be concluded that the core components are ivy saponin,pistil isoflavone,stigmasterol,sitosterol,clover terpene,(24S)-24-propylcholesterol-5-en-3 β-Alcohol,isorhamnetin,kaempferol,quercetin,deoxychlorothyroxine.The core target is serine/threonine protein kinase 1,tumor necrosis factor,interleukin 6,epidermal growth factor receptor,non-receptor tyrosine kinase,signal transduction and transcription activation factor 3,mitogen-activated protein kinase 3,heat shock protein,avian sarcoma protein gene(JUN)cystatin 3.According to GO analysis and KEGG analysis,biological processes mainly involve positive regulation of RNA polymerase II promoter transcription,positive regulation of gene expression,positive regulation of apoptosis process,protein phosphorylation and other reactions.Cell components mainly involve cell components such as nucleus,cell matrix,cytoplasm,nucleoplasm,cell plasma membrane,etc.The molecular function mainly involves protein binding,same protein binding,ATP binding,enzyme binding,transcription factor binding,protein kinase activity,protein serine/threonine kinase activity,etc.The signal pathway mainly involves PI3K-Akt signal pathway,cancer pathway,chemical carcinogenesis-receptor activation and other signal pathways.Conclusion:1.Through data mining of clinical literature,it can be concluded that the core traditional Chinese medicine compound "Huangqi-Rehmannia Preparata-Schisandra chinensis" is the core traditional Chinese medicine compound for the treatment of COPD by the golden and water mutually reinforcing method.2.Through the analysis of "Astragalus-Schisandra chinensis-Radix Rehmanniae Preparata" "The network pharmacology results of traditional Chinese medicine in the treatment of chronic obstructive pulmonary disease can be obtained from the core active components of the traditional Chinese medicine compound,such as sinomenin,calyx isoflavone,stigmasterol,sitosterol,isorhamnetin,kaempferol,and its key targets are AKT1,TNF,IL6,EGFR,SRC,STAT3,CASP3.Through GO analysis and KEGG analysis,it can be concluded that the traditional Chinese medicine compound can regulate the proliferation of cells by regulating the intervention protein kinase through the above core targets,Migration,apoptosis,and through regulating a series of signal pathways with PI3K-Ak T signal pathway as the core,to reduce inflammation,inhibit the secretion of airway mucus and airway remodeling. |
