Purpose:Through data mining,this study collected and sorted out the literature on the treatment of bronchial asthma with the method of “the generation between the metal and water”by contemporary physicians in the past 20 years,obtained the prescriptions used,analyzed and summarized the rules of prescription.Using the method of network pharmacology,we screened the effective components of the core drug pair for the treatment of bronchial asthma with the method of “the generation between the metal and water” mutually reinforcing,constructed the molecular network,and explored its potential mechanism of action in the treatment of bronchial asthma.Material and method:By searching the literature on the treatment of bronchial asthma with the method of “the generation between the metal and water” phase growth from January1,2000 to November 30,2022 on CNKI,Wanfang Data and VIP,we screened the literature according to the inclusion criteria,obtained the prescription,imported the drug into Excel and established a database to make statistics on the frequency,classification,nature,taste and meridian of the drug.SPSS Statistics 22.0 software was used for cluster analysis of drugs,and SPSS Modeler 18.0 software was used for association rule analysis of drugs.Through the TCMSP platform,the active components of the drug pair with the highest degree of support and its target were screened,and the gene name corresponding to the target protein was found through the Uniprot database.The "TCM-component-target network diagram" was constructed by using Cytascape 3.9.1 software.Use Gene Cards database,OMIM database and Dis Gent database to search the disease target of bronchial asthma,build the same target of core drug pair and bronchial asthma disease,use String platform to build PPI network model,and use Cytascape 3.9.1 software to optimize the PPI network model.Common genes were imported into David database for GO and KEGG enrichment analysis.Results:1.Data mining results: 197 prescriptions of traditional Chinese medicine were included in this study,involving 196 kinds of traditional Chinese medicine,with a total frequency of 2463 times.The ten traditional Chinese medicines with the highest frequency were astragalus,licorice,schisandra chinensis,pinellia ternata,earthworm,prepared rehmannia,poria cocos,cornus officinalis,epimedium and bitter almond.The most frequently occurring type of traditional Chinese medicine is deficiency tonic.The lung,spleen and kidney meridians ranked the top three.In the rank of drug properties,warm drugs appear the most,and sweet drugs appear the most.The results of cluster analysis showed that there were Gather 1 kinds: Rehmannia glutinosa,Poria cocos,Chinese yam,Cornus officinalis,Ophiopogon japonicus,Codonopsis pilosula;Gather 2 kinds: licorice,orange peel,angelica,pinellia;Gather 3 categories: astragalus,atractylodes macrocephala,schisandra chinensis,and perilla frutescens; Gather 4 categories: earthworm,bitter almond,gecko and epimedium.The association rule results showed that the drug pair with the highest support was Astragalus-Epimedium.2.Network pharmacology results: 40 active components of Epimedium and Astragalus were obtained through TCMSP database,corresponding to 230 targets.Through network topology analysis,the top three active compounds ranked highest were quercetin,kaempferol and luteolin.Through searching on Gene Cards database,OMIM database and Dis Gent database,3554 disease targets of bronchial asthma were finally obtained.Take the intersection of the two and get 172 drug and disease intersection targets.After constructing the PPI network,the core targets are AKT1,IL-6,TNF,VEGFA,JUN,IL-1β、 CASP3、EGFR、PTGS2、ESR1。The results of GO enrichment analysis showed that the action of astragalus membranaceus and epimedium on bronchial asthma involved 828 biological processes,88 cell components and 155 molecular functions.The results of KEGG enrichment analysis showed that there were 177 pathways involved in the treatment of bronchial asthma by astragalus membranaceus and epimedium,including tumor necrosis factor signal pathway,PI3K-Akt signal pathway,IL-17 signal pathway,etc.Conclusion:1.The method of the generation between the metal and water is mostly used to treat bronchial asthma in remission stage,and commonly used Chinese medicines such as Radix Astragali,Radix Glycyrrhizae,Fructus Schisandrae Chinensis,Rhizoma Pinelliae,Lumbricus,Radix Rehmanniae Preparata,Poria,Cornus officinalis,Herba Epimedii,and Semen Armeniacae Amarum.Drugs are mostly used to tonify deficiency,mainly to tonify lung qi and kidney yang,and enter the lung,spleen,kidney and liver meridians in the meridian.The drugs are most warm in nature and most sweet in taste,giving consideration to pungent dispersion and bitter discharge,which embodies the basic pathogenesis of bronchial asthma,not only tonifying lung and kidney,but also paying attention to strengthening spleen,fundamentally eliminating the generation of phlegm,eliminating the root of asthma,and at the same time cooperating with dispersing lung and relieving asthma.When treating this disease with the method of the generation between the metal and water,we should pay attention to the differentiation of lung and kidney qi deficiency,yin deficiency or yang deficiency,and whether there is phlegm and blood stasis,and treat it according to the patient’s condition.2.Based on the method of network pharmacology,the potential mechanism of astragalus membranaceus and epimedium in treating bronchial asthma is obtained: the main components of core drugs include quercetin,kaempferol,luteolin,etc.,acting on AKT1,IL-6,TNF,VEGFA,JUN,IL-1β,CASP3,EGFR,PTGS2,ESR1 and other targets are used to treat bronchial asthma by regulating signal pathways such as interleukin-17,tumor necrosis factor,PI3K-Akt and so on. |
