Purpose: The data mining technology was used to analyze the medication patterns in clinical literature about the treatment of chronic obstructive pulmonary disease(COPD)with oral Chinese medicine in the past 20 years.The drug pairs with the highest degree of association rules were selected and the action mechanisms and potential targets in the treatment of chronic obstructive pulmonary disease(COPD)were analyzed by network pharmacology,so as to provide reference for the experimental research direction of TCM treatment for COPD(lung-kidney qi deficiency syndrome)and the development of new drugs.Method: The relevant literature on the clinical treatment of chronic obstructive pulmonary disease(lung-kidney qi deficiency syndrome)in traditional Chinese medicine was collected and screened through the literature databases of CNKI,VIP,Wanfang,pub med,and others.The COPD database was constructed,and the data information of traditional Chinese medicine was imported into Excel.SPSS Modeler18.0 and SPSS Statistics were used as data mining tools for frequency,association and clustering analysis to explore the therapeutic principles and treatment,as well as the law of drug compatibility and formula.The drug pair(group)with the highest correlation analysis degree was analyzed for potential targets and possible action mechanism by network pharmacology.Results:1.A total of 32 prescriptions were screened and included,involving a total of 71 Chinese medicinals.A total of 15 Chinese medicinals with the frequency of more than six times were selected,which were Radix Astragali seu Hedysari,Fructus Schisandrae Chinensis,and Radix Rehmanniae Preparata in descending order.In the drug classification,the reinforcing drugs were the most frequently used,followed by the drugs for relieving cough,eliminating phlegm and relieving asthma,and the drugs for regulating qi,resolving stasis and inducing diuresis were the second most frequently used.Drugs with moderate temperature in the four qi meridians had the highest frequency.Among the five flavors,sweet flavor was used most frequently,followed by pungent flavor.The frequency of entering lung meridian is the highest,followed by kidney meridian.2.Based on the analysis of association rules and the one with the highest association rule was Radix Asteris–Radix Astragali seu Hedysari.3.Systematic clustering analysis was performed on high-frequency drugs,and the results showed that they were divided into three categories.The first category was Aster,Farfara,Perilla,Radix Astragali,Radix Codonopsis,Radix Glycyrrhizae,Rhizoma Pinelliae,fructus schisandrae chinensis and radix rehmanniae preparata.The second type is Poria,rhizoma Atractylodis Macrocephalae,herba Epimedii,pericarpium Citri Reticulatae,and radix Salviae Miltiorrhizae;The third type is gecko.4.There were 36 target compounds of key drug pairs.After analysis,we found that 26 potentially effective active compounds were quercetin,kaempferol,and β-sitosterol.A total of 203 intersecting targets between COPD(lung-kidney-qi deficiency syndrome)and core drug pairs and 6 core targets located in protein interaction were screened,which were AKT1,JUN,MAPK1,TP53,TNF and RELA,respectively.Through GO functional enrichment analysis of core targets and KEGG pathway enrichment analysis,it can be preliminarily predicted that the functional pathways involved in relevant targets are oxygen level response,reactive oxygen species response,steroid hormone response,etc.A variety of signaling pathways such as AGE-RAGE signaling pathway,anti-viral infection(Hepatitis B,Measles),PI3K-Akt signaling pathway and NF-k B signaling pathway may be the relevant pathways for the core drug pairs to act on COPD(lung-kidney qi deficiency syndrome).Conclusion:1.COPD(qi deficiency of lung and kidney)is caused by deficiency in nature and excess in superficiality,with strengthening vital qi as the main principle of treatment.It indicated that tonifying qi and eliminating phlegm and blood stasis were the key.Medication is characterized by warmth,and the majority of the herbs are sweet and punge nt in taste.The treatment is based on lung and kidney.The drug combinations obtained from correlation analysis and clustering analysis have the characteristics of treating both the manifestation and root cause of the disease,which can be used for reference in clinical formulation.2.The 26 active components in Radix Asteris-Radix Astragali seu Hedysari play the roles of anti-inflammation,anti-oxidation,reducing lung tissue damage,inhibiting airway remodeling and airway mucus hypersecretion in the treatment of COPD(lung-kidney qi deficiency syndrome),by regulating key targets such as AKT1,JUN,MAPK1,TP53, TNF,as well as action pathways such as AGE-RAGE signaling pathway,anti-viral in fection(Hepatitis B,Measles),PI3K-Akt signaling pathway and NF-k B signaling path way.
