| Objective In this study,the pollen allergens of Artemisia annual and Artemisia sieversia na in children in Hohhot were screened and biological information was analyzed.To pr ovide basic data for understanding the sensitization characteristics and related mechanis ms of serum specific Ig E(s Ig E)antibodies to two artemisia pollen in children with Aller gic Rhinitis(AR)in this area.To provide basis for accurate diagnosis and immunothera py of allergic diseases.Methods According to the inclusion and exclusion criteria,the cases and serum of childr en who were clinically diagnosed with AR and tested positive for s Ig E antibody of Arte misia allergen were collected.The pollen of A.annua and A.sieversiana was collected,and the proteins were extracted.The serum of the children was used as the primary antib ody.The potential allergens were screened by western blot,and qualitative analysis was performed using shotgun method.Results(1)The bands of Relative molecular mass(M r)of 12,25 and 70 KDa of two artemisia pollen extracts were commonand,there was dif ference in the positive reaction of the bands between them.Positive reaction bands were detected in serum of 27 allergic children with certain differences,including 14 cases wit h positive bands at Mr 70 KDa of both pollen extracts,10 cases with positive bands at M r 25 KDa of both pollen extracts,and 3 cases with positive bands at Mr 12 KDa of both p ollen extracts.In addition,9 cases only had positive bands on Mr 70 KDa of Artemisia a nnua pollen extract,and 1 case only had positive bands on Mr 12 KDa of one of the polle n extracts,respectively.The control children were negative.(2)The positive rate of Arte misia annua and Artemisia sieversiana allergy was 100% and 74.07%,respectively.The degree of binding between Artemisia annua pollen extract and serum s Ig E antibody of c hildren in this area was higher than that of Artemisia sieversiana.(3)The Mr 12,25 and 70 KDa allergens in Artemisia annua pollen extract were identified as Art an 3,Art an 25 KDa and Art an 7,respectively.The Mr 12,25 and 70 KDa allergens in artemisia siever siana pollen extract were Art si 3,Art si 25 KDa and Art si 7,respectively.(4)The positiv e rates of Art an 3,Art an 25 KDa and Art an 7 were 14.81%,37.04% and 85.19%,respe ctively.The positive rates of Art si 3,Art si 25 KDa and Art si 7 were 14.81%,37.04% a nd 51.85%.Art an 3 and Art si 3 had no significant difference in sensitization,nor did Art an 25 KDa and Art si 25 KDa,while Art an 7 and Art si 7 had significant difference.(5)Art an 7 and Art si 7 allergenic difference is obvious,according to biological informa tion than two plant sources of antigen peptides loci exist obvious difference sequence of amino acids,it may be lead to the same children in Mr 70 KDa only show the positive re sponse to A.annua stripe,and no synchronous positive reaction to A.sieversiana.Con clusion(1)The pollen of A.annua and A.sieversiana showed similar allergen profiles,a nd the main allergen molecules causing AR in children in Hohhot were Mr 12,25 and 70 KDa pair of strain allergens.Art an 3/Art si 3,Art an 25KDa/Art si 25 KDa and Art an7/Art si 7 respectively.(2)There were differences in the sensitization of the same serum t o the same location of two plants,especially in the Mr 70 KDa band,the sensitization rat e of A.annua was higher than that of A.sieversiana.The biological information compari son between the two allergens(Art an 7,Art si 7)sequence sites showed obvious differe nces.(3)The serum s Ig E antibody recognition of A.annua pollen extract and allergic chil dren in Hohhot was higher than that of A.sieversiana. |
