Purpose:Allergic rhinitis is an allergic disease that occurs in the nose,and its pathogenesis is still not very clear.Recent studies have shown that the occurrence of allergic rhinitis is related to genetic factors and is a group of diseases regulated by multiple genes.In this experiment,we analyzed the gene expression profiles of patients with Artemisia pollen sensitization,dust mite sensitization,and non-allergic rhinitis to find out whether there are differentially expressed and significantly differentially expressed genes,and explore the sensitization of significantly differentially expressed genes in Artemisia.The mechanism of action in the pathogenesis of allergic rhinitis sensitized by dust mites.Methods: Uni CAP system detects and groups: Group A is AR patients sensitized by Artemisia pollen;Group B is AR patients sensitized by dust mite,and Group C is non-AR patients with no clinical symptoms and negative test.Collect 3ml of peripheral blood from three groups A,B and C,extract total RNA and send it to the company for mRNA sequencing and differential expression analysis.Results: 1.Compared with the non-allergic rhinitis group,the Artemisia pollen group has 140 genes significantly differently expressed,97 of which are up-regulated and43 down-regulated;the Artemisia pollen group has 727 significantly differently expressed genes compared with the dust mite group.Among them,212 were up-regulated and 515 were down-regulated.Compared with the non-allergic rhinitis group,the dust mite group had 224 differentially expressed genes,of which 170 were up-regulated and54 were down-regulated.2.The up-regulated genes are mainly CXCR4,CXCL8,CNR2,PTGDR2,and the down-regulated genes are mainly HCAR2.The up-regulated genes are mainly involved in inflammation,chemokine signaling pathways,and G protein-coupled receptor signaling pathways.Down-regulated genes are mainly involved Apoptosis pathway.Conclusions:1.There are significant differences in mRNA gene expression profiles in AR patients sensitized by Artemisia pollen,AR patients sensitized by dust mites,and non-AR patients.2.CXCR4,CXCL8,CNR2,PTGDR2,HCAR2 are significantly differentially expressed in AR patients sensitized by Artemisia pollen,suggesting that they may be involved in the pathogenesis of Artemisia pollen sensitized AR. |
