| Hippophae is a genus of sea buckthorn in the family Hojicaceae,which is highly adaptable and is the tree of choice for windbreak and sand-fixing forests.Its roots,stems,leaves,fruits and seeds contain a large number of nutrients and biologically active substances and are of great economic value.With the increased awareness of the value of sea buckthorn,the demand for it has also increased,making the rapid breeding of sea buckthorn seedlings with good traits an urgent problem.The aim of this study was to analyse the advantages of developing Central Asian sea buckthorn by investigating the comprehensive traits of Central Asian sea buckthorn and Mongolian sea buckthorn;and to establish a fast breeding system using tissue culture as a technique by studying the various stages of priming,succession,rooting and domestication and transplanting using the preferred Central Asian sea buckthorn varieties WS-1 and WS-2 as experimental materials.The results of the study are as follows:(1)Through the investigation of the comprehensive characteristics of Central Asian sea buckthorn and Mongolian sea buckthorn,it was concluded that the fruit characteristics of Central Asian sea buckthorn are inferior to those of Mongolian sea buckthorn and its economic value is not as good as that of Mongolian sea buckthorn,but in terms of biology Central Asian sea buckthorn has higher growth capacity and stronger resistance to adversity,and is more valuable for improving barren land.Therefore,it is more advantageous to develop Central Asian sea buckthorn for the purpose of environmental improvement.(2)The best time for in vitro culture of WS-1 and WS-2 was found to be May for both varieties of field bud selection and February for hydroponic buds;the best disinfection method was 0.1% Hg/L for 3min + 75% alcohol for 10 s.The optimum hormone combination for the induction of adventitious buds of WS-1 was 1/2MS + 0.4 mg/L 6-BA,with an induction rate of 93.33%;for stem tip succession proliferation The optimum medium for stem tip succession proliferation was 1/2MS+0.2mg/L 6-BA,with a proliferation factor of 1.77;the optimum medium for stem segment succession proliferation was 1/2MS+0.5mg/L 6-BA,with a proliferation factor of 2.56;the optimum medium for healing tissue succession proliferation was1/2MS+0.5mg/L 6-BA,with a 92.22% induction rate of adventitious shoots;the optimum medium for rooting sterile seedlings was 1 The optimum hormone combination for WS-2 adventitious shoot induction was 1/2MS+0.5mg/L 6-BA+0.5mg/L IAA with 87.78% induction rate;the optimum medium for stem tip succession proliferation was 1/2MS+0.5mg/L 6-BA with a proliferation factor of 1.78;the optimum medium for stem segment succession proliferation was 1/2MS+0.5mg/L 6-BA with a proliferation factor of1.78;the optimum medium for stem segment succession proliferation was 1/2MS+0.5mg/L 6-BA with a proliferation factor of 1.78;the optimum medium for stem segment succession proliferation was1/2MS+0.5mg/L 6-BA with a proliferation factor of 1.78.The most suitable medium for successive proliferation of stem segments was 1/2MS + 0.5mg/L 6-BA + 0.3mg/L IAA,with a proliferation factor of2.33;the most suitable medium for successive proliferation of healing tissue was 1/2MS + 0.5mg/L 6-BA +0.5mg/L IAA,with a 91.11% induction rate of adventitious buds;the best medium for rooting sterile seedlings was 1/2MS + 0.1mg/L 6-BA + The best rooting medium was 1/2MS+0.1mg/L 6-BA+1.0mg/L IBA,and the rooting rate was 93.33%.The best hormone treatment time for rooting outside the bottle was10 s for both species,and the rooting rate was 96.67% and 95.56% respectively.The best substrate for transplanting of both species was 1:2 of grass charcoal soil and perlite,and the survival rate was 95.56%and 93.33% respectively. |
