As a new fruit native to China,kiwifruit has been commercialized in China for nearly 40 years.The rapid development in recent years,the Chinese kiwi fruit industry,both cultivation area and yield are one of the world's first,commercialization degree deepening,but lack of high quality seedlings,and extensive seedling technique seriously restricts the development of Chinese kiwifruit industry.In order to further commercialize,scale and internationalize the kiwifruit industry,it is necessary to reform and develop the seedling.Based on tissue culture seedling period is short,without being limited by the growing season,showed characteristics of nursery stock and tidy,this experiment mainly study kiwifruit seed to establish the system of rapid propagation,especially for kiwi several links of roots,refined seedling transplanting experiment research,the main varieties are ‘Qi Hong',‘Nongda Yuxiang',‘Actinidia macrosperma'and‘X-1',all the materials collected from the northwest agriculture and forestry university of science and technology of kiwi fruit test station,adopt the varieties of crown and band as the explant tissue culture shoots stem section to in vitro culture of each type,the kiwi fruit to create the optimal explant each type sterilization processing time,The main experimental results are as follows:1.The best sterilization process of various types of explants: first,75% alcohol disinfection for 15 seconds,sterile water rinsing 3 times,and then 0.1% hg sterilization(time depending on variety),sterile water rinsing 5 times.The sterilization time of each variety 0.1 was 4min,3min,4min,5min and 7min respectively.The corresponding survival rates were 45%,65%,30%,50% and 75% respectively.2.The optimum primary culture medium of each variety: ‘Qi Hong' was MS+30g/L sucrose +6.8g/L AGAR +3mg/L 6-ba +0.2mg/L NAA,‘Nongda Yuxiang'was 73.33%;The growth rate was 46.67%,and the germination rate was 46.67%.The‘Actinidia macrosperma' was MS+30g/L sucrose +6.8g/L AGAR +2mg/L 6-BA+0.2mg/L NAA,the germination rate was 76.67%.‘X-1' was MS+30g/L sucrose +6.8g/L AGAR +1mg/L ZT,and the germination rate was 63.33%.3.The best secondary multiplication culture medium of each variety: ‘Qi Hong' was MS+30g/L sucrose +6.8g/L AGAR +3mg/L 6-ba +0.15mg/L NAA+0.15mg/L GA3,the proliferation coefficient was 4.5,and the average height was 3.53 cm.‘Nongda Yuxiang' was 4.75,and the average plant height was 3.83 cm.‘Actinidia macrosperma' was MS+30g/L sucrose +6.8g/L AGAR +2mg/L 6-BA+0.05mg/L NAA,the proliferation coefficient was 4.25,and the average height was 4.45 cm.‘X-1' was MS+30g/L sucrose +6.8g/L AGAR +1.5mg/L ZT,the proliferation coefficient was 6.02,and the average height was 3.2cm.4.The best bottles for each type and keep rooting medium: ‘Qi Hong' was 1/2 MS + 25 g/L + 6.8 g/L AGAR + 0.6 mg/L IBA + 0.30.6 mg/L IAA,began to take root for 11 days,the average rooting rate is 70%,average root number 5.37,the average root length 4.18 cm,smaller callus;‘Nongda Yuxiang' was 1/2MS+25g/L sucrose +6.8g/L AGAR +0.6mg/L IBA,and the rooting time was the 11 th day,the average rooting rate was 57.5%,the average rooting number was 5.63,the average root length was 3.42 cm,the more the injury tissue was smaller.‘Actinidia macrisperma' was 1/2MS+25g/L sucrose +6.8g/L AGAR +0.2mg/L IBA,and the rooting time was 7 days,the average rooting rate was 70.01%,the average rooting number was 8.75,the average root length was 4.5cm,and the healing tissue was small.The optimum culture medium for ‘X-1' was 1/2MS+25g/L,sucrose +6.8g/L AGAR +0.6mg/L IBA+0.1mg/L IAA,the rooting time was 8 days,the average rooting rate was 76.25%,the average rooting number was 8.5,and the average root length was 3.1cm,and the healing tissue was smaller.5.Each type autotrophic best growth regulator levels: ‘Qi Hong' and ‘Nongda Yuxiang' 0.3 mg/L ABT 1,‘Actinidia macrosperma' and ‘X-1' is 0.5 mg/L ABT 1,the roots of time respectively,5d,6d,6d,6d,the average rooting rate was 83.75%,87.55,88.25%,89%,the average number of roots were 12.5,14.25,6,6.25;The average root length was 3.34 cm,3.37 cm,3.29 cm,and 3.33 cm.6.The best way to prepare the seedlings:put in autotrophic root box of smelting plants,the highest survival rate of seedlings during smelting,7d after ‘Qihong' and‘Nongda Yuxiang' good seedling rate can reach 97%,‘Actinidia macrosperma' and ‘X-1' good seedling rate can reach 100%,the half-open lid miao times,7 d after‘Qi Hong' and ‘Nongda Yuxiang' good seedling rate can reach 89.1%,‘Actinidia macrosperma' and ‘X-1' good seedling rate can reach 92.4%.7.The best transplanting matrix: straw carbon and rural soil were matched with a ratio of 3:7,and the survival rate was highest after transplanting 45 d.The ‘Qi Hong' and ‘Nongda Yuxiang' could reach 92.4%,and ‘Actinidia macrosperma' and ‘X-1' could reach 85.8%.8.Best transplanting root length: ‘Qi Hong' and ‘Nongda Yuxiang',the root length is the highest in 1cm~4cm,and the survival rate can reach 100% after transplanting 25 d.For ‘Actinidia macrosperma' and ‘X-1',the root length was 2.5cm~4cm,the survival rate was the highest,and the survival rate after 15 d was 100%.
