ObjectiveThe patients with Type 2 diabetes mellitus(T2DM)are getting younger and younger, and various complications of diabetes are becoming more common.Non-alcoholic fatty liver disease(NAFLD)is a disease caused by fatty degeneration of liver cells. And it is a common complication of T2DM.T2DM and NAFLD are mutually causal and mutually promote each other, and become a global health problem that needs to be solved urgently.Asprosin is a cytokine discovered in recent studies. And it is closely related to T2DM and NAFLD,and plays a very important role in the entire pathogenesis. But there are few studies on the relationship between asprosin and T2DM combined with NAFLD at home and abroad. This article aims to explore the relationship between asprosin and T2DM combined with NAFLD,and to find the related factors of that.It may help the scientists to treat the T2DM combined with NAFLD.MethodThe 150 patients and health examiners who visited the hospital from November2019 to November 2020 were selected and divided into three groups.T2DM combined with NAFLD(T2DM +NAFLD group)50 cases(28 males,22 females),pure T2DM patients(T2DM group)50 cases(23 males,27 females),normal people(NC group)50cases(24 males,26 females)were selected. The fasting blood of the patients were collected, and detected the following indicators.Fasting insulin(Fins),Fasting blood sugar(FBG),Fasting C peptide(C peptide),Glycated hemoglobin(Hb A1c);Low density lipoprotein cholesterol(LDL-C),high density lipoprotein cholesterol(HDL-C),triglyceride(TG),total cholesterol(TC);aspertate aminotransferase(AST),glutamate aminotransferase(ALT),glutamyl transpeptidase(GGT);Uric acid(UA),Urea(Ur),Creatinine(Cr).Waist-hip ratio(WHR),Body Mass Index(BMI),Beta cell function index(HOMA-β),Insulin resistance index(HOMA-IR)were calculated. The blood samples(×3000r/min)were centrifuged for 10 minutes. The serum samples were collected and stored in a refrigerator at-80°C.The enzyme-linked immunosorbent(ELISA)method was used to detect the concentration of asprosin.Result1.Compared with the NC group, asprosin,BMI,waist, hip,FBG,Hb A1 c,Fins,HOMA-IR,GGT,TG,TC,LDL-C in the T2DM group increased,C peptide,HOMA-β,HDL-C decreased, the difference was statistically significant(P<0.05).2.Compared with the NC group, asprosin,BMI,waist, hip,FBG,Hb A1 c,Fins,HOMA-IR,ALT,GGT,TG,TC,LDL-C in the T2DM+NAFLD group increased,Cpeptide,HOMA-β,HDL-C decreased,he difference was statistically significant(P<0.05).3.Compared with the T2DM group, asprosin,BMI,waist, hip,FBG,Hb A1 c,Fins,HOMA-IR,ALT,GGT,TG in the T2DM+NAFLD group increased,C peptide and HOMA-β decreased,he difference was statistically significant(P<0.05);TC,LDL-C increased,HDL-C decreased had no significant difference(P>0.05).4.The level of serum asprosin are positively correlated with BMI,waist, hip,FBG,Hb A1 c,HOMA-IR,GGT,ALT,TC,TG,LDL-C(P<0.05).And negatively correlated with C peptide and HDL-C(P<0.05).While there was no correlation with height,weight,WHR,AST,Ur,Cr,UA(P>0.05).5.BMI(β=7.648,P=0.008)and HOMA-IR(β=3.342,P=0.010)are independently related factors of asprosin.6.There were no statistically differences in gender, age,WHR,AST,Ur,Cr,UA among the three groups(P>0.05).Conclusion1.The level of serum asprosin in NC group,T2DM group, and T2DM+NAFLD group increased gradually. It can be used to assess the patient’s Pancreas function and the severity of fatty liver.2.The level of serum asprosin has relationship with BMI,waist, hip,FBG,Hb A1 c,HOMA-IR,GGT,ALT,TC,TG,LDL-C,C peptide,HDL-C.BMI,HOMA-IR are related independently factors of asprosin. The higher of serum asprosin level, the more severe IR,and the blood sugar level was higher.3.Asprosin can cause the deterioration of IR and accelerate the failure of β-cell function.Lowering the level of serum asprosin can improve IR and lower blood sugar levels. Therefore, asprosin can be used as a new target for the treatment of T2DM and NAFLD.
