A Study On Serum Asprosin Levels In Patients With Nonalcoholic Fatty Liver Disease

Objective:Non-alcoholic fatty liver disease(NAFLD)is closely related to insulin resistance and diabetes.At present,the clinical diagnosis of NAFLD and the understanding of the severity of the gold standard are still the liver biopsy,and it remains the lack of effective treatment measures.The aim of this study was to study the changes of the level of serum Asprosin in patients with nonalcoholic fatty liver disease and/or type 2diabetes mellitus(T2DM).To discuss the value of Asprosin on diagnosis of NAFLD and/or T2 DM,and provide a new target for the clinical treatment of NAFLD and/or T2 DM.Methods:1.From July 2017 to March 2018,200 outpatients and hospitalized patients in the First People’s Hospital of Jiujiang were enrolled,including 50 healthy people,50 NAFLD patients,50 patients with NAFLD and T2 DM,and 50 patients with T2 DM.The actual participants were 50 healthy people,44 NAFLD,58 NAFLD and T2 DM,and 32T2 DM.They were matched in terms of age,sex,body mass index(BMI)and education.Fasting blood was sampled.The levels of serum Asprosin and adiponectin were determined by ELISA.Fasting serum glucose(FSG),glycated hemoglobin A1c(HbA1c),blood uric acid(UA),serum total cholesterol(TC),triglyceride(TG),alanine amiotransferase(ALT),aspartate aminotransferase(AST),high density lipoprotein C(HDL-C),low density lipoprotein(LDL-C),and fasting serum insulin(FINS)were detected by automatic biochemistry analyzer.NAFLD was diagnosed by B ultrasound,and NFS score was used to evaluate the severity of NAFLD.Serum Asprosin levels in the four groups was compared.According to the severity of NAFLD,NAFLDand NAFLD + T2 DM patients were divided into three groups,the light,medium and severe,respectively.The lever of Asprosin in the three groups were discussed.On the other hand,according to the NFS(NAFLD Fibrosis Score)(no fibrosis to fibrosis grade 2: <-1.455;fibrosis degree was uncertain:-1.455~0.675;fibrosis 3~4 level: >0.675),NAFLD and NAFLD + T2 DM patients were evaluated.All the patients were divided into three groups,and the differences of Asprosin between the three groups were discussed.2.SPSS ver 20 and GraphPad Prism 6.0 software were performed for all statistical analyses.statistical Kolmogorov-Smirnov test data is normal distribution.Normal distribution of data to X ± s said that the non-normal distribution of data,its natural logarithm normalized data analysis before the next step.Two sample comparing with independent samples t test.A single factor variance analysis for the comparison of the diversity of the average numbers.The non-normal distribution data is represented by median(four quantile)[M(P25,P75)].Group design of two samples is compared with non-parametric Mann-Whitney rank sum test.The correlation between the two groups was analyzed by Pearson analysis.P value < 0.05(2-side)was considered to be statistically significant.Results:1.The serum Asprosin levels in NAFLD,T2 DM and T2 DM + NAFLD were significantly higher than those normal group(p <0.01),but serum adiponectin was significantly decreased(p <0.001).2.In group NAFLD,a linear correlation was showed in PTL,FSG,TG,serum albumin,NFS with Asprosin,,respectively,p value was 0.015,0.004,0.025,0.007,0.009,respectively.r value was 0.3653,-0.4304,0.3383,0.4124,-0.3945,respectively.Negative correlation was showed both in FSG and NFS with Asprosin.Positive correlation was showed among PLT,TG and ALB with Asprosin.3.In the T2 DM group,linear and positive correlation was showed in PTL,FSG,TG,TC and LDL-C with Asprosin,respectively.p values was 0.002,0.015,0.004,0.001,0,respectively.And r value was 0.5256,0.4266,0.4963,0.5708,0.5986,respectively.4、In T2 DM + NAFLD group,the level of Asprosin was linearly correlated with PTL,FSG,TG,ALB,and HOMA-IR.The p values were(0.007,0.019,0.004,0.027,0.003),r values were(0.3506,0.3069,0.3757,0.2902,0.3972),respectively.Asprosin and NFS were linear,and negatively correlated(p=0.010;r=0.3342).Conclusions:1.Compared with control,the level of Asprosin in NAFLD patients was significantly increased.2.Asprosin was an independent risk factor for NAFLD combined with T2 DM.3.In NAFLD patients,regardless of T2 DM,the NFS was negatively correlated with Asprosin.The detection of serum Asprosin had certain guiding significance for determining the severity of liver fibrosis.
