Objectives Through the establishment of a mouse model of chronic unpredictable mild and stimulus(CUMS)depression,the effects of huazhuojiedu shugan formula(HJSF)on the emotional behavior and the expression of β-endorphins(β-EP)in the hypothalamus of mice model of depression were observed,so as to provide theoretical basis and guidance for the diagnosis and treatment of clinical depression.Methods 54 male clean grade ICR mice were randomly divided into blank control group,model group,HJSF low dose group,HJSF medium dose group,HJSF high dose group and fluoxetine group.With the exception of the blank control group,mice in all the other groups continuously replicated the depression model using the method of chronic unpredictable mild stress for 21 days in total.After the model of depression was copied,the mice were determined to determine whether the model was successful or not based on the results of sucrose preference test.After the model was successfully constructed,the HJSF group was treated with the HJSF through gavage,the fluoxetine group was treated with the fluoxetine through gavage,the blank control group and the model group were treated with normal saline through gavage for 21 consecutive days.Then the novelsuppressed fedding test,tail suspension test,the forced swimming test,the elevated plus maze test and so on were carried out to test the emotional behavior.Immunohistochemistry was used to detect the expression level of β-EP in the dorsomedial hypothalamic nucleus(DMH)and ventromedial hypothalamic nucleus(VMH)in depressed mice.All data were statistically processed using SPSS 20.0 software,the comparison of the data conforming to the normal distribution and homogeneous variance among the groups was performed by one-way anova,and the non-parametric test was performed for variance heterogeneity.P<0.05 was considered to be significant.Results 1)sucrose preference test: before modeling,there was no significant difference in the sugar water preference ratio of each group(P>0.05).After modeling,compared with the blank control group,the sugar water preference ratio of the other groups decreased,with a significant difference(P<0.01).After treatment,compared with the blank control group,the sucrose preference ratio of mice in the model group was reduced,and the difference was highly significant(P<0.01).Compared with the model group,the sucrose preference ratio of mice in the HJSF medium dose group,high dose group and the fluoxetine group were all increased,and the difference was statistically significant(P<0.01).Compared with the fluoxetine group,the sucrose preference ratio of mice in the HJSF medium and high dose groups had no significant difference(P>0.05).2)novelsuppressed fedding test: compared with the blank group,the latency of novel-suppressed fedding test was prolonged in the model group,with a significant difference(P<0.01).Compared with the model group,the latency of novel-suppressed fedding test in the HJSF medium dose group,high dose group and the fluoxetine group were shortened,and the difference was significant(P<0.01).Compared with the fluoxetine group,there was no significant difference in the latency of novel-suppressed fedding test in mice with HJSF medium and high dose groups(P>0.05).3)tail suspension test: compared with the blank group,the total tail suspension time of mice in the model group was prolonged,the difference was significant(P<0.05).Compared with the model group,the total suspended tail immobility time of mice in each treatment group was reduced,and the difference was significant(P<0.05).Compared with the fluoxetine group,the total suspended tail immobility time of the mice in the HJSF medium and high dose groups showed no significant difference(P>0.05).4)forced swimming test: compared with the blank group,the total immobility of mice in the model group was increased,with significant difference(P<0.01).Compared with the model group,the total immobility time of mice in each treatment group was reduced,with significant differences(P<0.05).Compared with the fluoxetine group,the total immobility time of forced swimming in the HJSF medium and high dose groups showed no significant difference(P>0.05).5)elevated plus maze test: compared with the blank group,the distance and time in open arms of mice in the model group were reduced,and the difference was significant(P<0.05).Compared with the model group,the distance and time in open arms of mice in the HJSF medium dose group,high dose group and the fluoxetine group were increased,and the difference was significant(P<0.05).Compared with the fluoxetine group,there was no significant difference in the distance and time in open arms of mice in the HJSF medium and high dose groups(P>0.05).Immunohistochemical determination: compared with the blank group,expression of β-endorphins in DMH and VMH in model was reduced,with significant difference(P<0.01);Compared with the model group,the expression of β-endorphins in DMH and VMH in each treatment group was increased,and the differences were significant(P<0.05).Compared with the fluoxetine group,there was no significant difference in the expression of β-endorphins in DMH and VMH in the HJSF medium and high dose groups(P>0.05).Conclusions 1 The results of the experimental detection of sucrose preference test in mice indicate that the depression model replicated by the CUMS method in this study is successful.2 Huazhuo jiedu shugan formula can effectively improve the CUMS pleasure lack of depression model mice and decrease environmental exploration desire in depression behavior and increase the expression of β-endorphin in DMH and VMH,suggests that the antidepressant mechanism may be related to higher the expression of beta-endorphin in DMH and VMH.3 The antidepressant effect of huazhuo jiedu shugan formula in medium and high doses groups is superior to that of huazhuo jiedu shugan formula
