Objective: To find the mechanism of action of Huazhuojiedu formula in the treatment of the CEG by observing the clinical efficacy of Huazhuojiedu formula in the treatment of the Chronic Erosive Gastritis patients with Zhuodu intrinsic and the expression of Raf-1/MEK-2/ERK-1 signaling pathway,and provide a theoretical basis for the treatment of chronic erosive gastritis patients by using Zhuodu theory.Methods: Seventy patients with CEG with Zhuodu intrinsic were randomly divided into treatment group(35 cases)and control group(35 cases).The treatment group was treated with Huazhuojiedu formula.The control group received oral A la tan Wu wei wan,and 3 months was a course of treatment.Both groups received 2 courses of continuous medication.The symptom scores,gastroscopy and pathological effects were observed before and after treatment.Immunohistochemistry and Western blot were used to observe the changes of Raf-1,MEK-2 and ERK-1 protein expression in gastric mucosa before and after treatment.Results:1.Before treatment,there was no significant difference in symptom scores,gastroscopy and pathological conditions between the two groups(P>0.05),which was comparable.2.Symptom score: Compared with before treatment,the symptom scores of the two groups were decreased after treatment,the difference was statistically significant(P<0.05),and the symptom score of the treatment group was lower than that of the control group,the difference was statistically significant(P<0.05).3.TCM syndrome efficacy: After treatment,the total effective rate was96.97%,17 cases were cured,8 cases were markedly effective,7 cases were effective,and 1 case was ineffective.The total effective rate of the control group was 60.61%,7 cases were cured,and 5 cases were markedly effective.In 8 cases,13 cases were ineffective.The treatment group was better than the control group,and the difference was statistically significant(P<0.05).4.Gastrointestinal effect: After treatment,the total effective rate was93.94%,14 cases were cured,7 cases were markedly effective,10 cases were effective,and 2 cases were ineffective.The total effective rate of the control group was 81.82%,5 cases were cured,8 cases were markedly effective,14 cases were effective,and 6 cases were ineffective.The treatment group was better than the control group,and the difference was statistically significant(P<0.05).5.Pathological effect: After treatment,the total effective rate was 81.82%,11 cases were cured,9 cases were markedly effective,7 cases were effective,and 6 cases were ineffective.The total effective rate of the control group was54.55%,6 cases were cured,5 cases were markedly effective,and 7 cases were effective,and 15 cases were ineffective.The treatment group was better than the control group,and the difference was statistically significant(P<0.05).6.Immunohistochemical results: Before treatment,there was no significant difference in the expression of Raf-1,MEK-2 and ERK-1 protein in gastric mucosa between the two groups(P>0.05),which was comparable.Compared with before treatment,the expression of Raf-1,MEK-2 and ERK-1 protein in gastric mucosa decreased after treatment,the difference was statistically significant(P<0.05),and the expression levels of Raf-1,MEK-2and ERK-1 in the gastric mucosa of the treatment group were significantly lower than those in the control group,and the difference was statistically significant(P<0.05).7.Western blot results: Before treatment,the relative expressions of Raf-1,MEK-2 and ERK-1 proteins in gastric mucosa of the two groups were not significantly different(P>0.05),which was comparable.Compared with before treatment,the relative expression of Raf-1,MEK-2 and ERK-1 protein in gastric mucosa decreased after treatment,the difference was statistically significant(P<0.05),and the relative expressions of Raf-1,MEK-2 and ERK-1 in gastric mucosa of the treatment group were lower than those in the control group,the difference was statistically significant(P<0.05).Conclusion:1.Huazhuo Jiedu formula can significantly improve the clinical symptoms,gastroscopy and pathology of patients with CEG with Zhuodu intrinsic.2.The mechanism of Huazhuo Jiedu formula in treating CEG with Zhuodu intrinsic may be related to down-regulation of Raf-1,MEK-2,ERK-1protein,inhibition of abnormal activation of Raf-1/MEK-2/ERK-1 signaling pathway and prevention of abnormal proliferation and differentiation of cells.
