Study On The Effect Of Sanshen Yizhi Capsule On The Learning And Memory Ability With Vascular Dementia And The Underlying Mechanism In Rats

Objective:To study the effect of Sanshen Yizhi Capsule on learning ability and memory with vascular dementia(VD)and its mechanism in rats.Method:The weight-matched SD rats the were randomly divided into 5 groups with 10 per group:sham operation group,VD group,Sanshen Yizhi capsule high-dose group,Sanshen Yizhi capsule low-dose group,and western medicine group(Nicergoline group).Except for the sham operation group,the other groups were induced to develop VD by using a 2-vessel blockade+sodium nitroprusside method.At the end of Sanshen Yizhi and Nicergoline administration,the learning ability and memory of rats was studied by Morris water maze assay,the pathological changes of hippocampus were studied by HE stain,and serum level of AchE,Bcl-2 and Bax were detected by enzyme-linked immunosorbent assay(ELISA).Results:1.The results of Morris water maze test showed that the ascend platform of latent period(APLP)of VD group was significantly longer than that of the sham operation group(P<0.05).Compared with VD group,the APLP of Sanshen Yizhi Capsule groups and the western medicine group significantly shortened.Compared with VD group,the APLP of low-dose Sanshen Yizhi Capsule was significantly shortened on the fourth day(P<0.01).The APLP of high-dose group was significantly shortened in the second and third days(P<0.05),which was further shortened on the fourth day(P<0.01).2.The results of HE staining showed that the cells in the hippocampus of rats in sham operation group were arranged closely and orderly,with a similar cell size and shape.The number of cells in the VD group was greatly reduced,the cells were arranged sparsely,and the cell morphology was different.Compared with VD group,the number of cells in the low-dose group,high-dose group of Sanshen Yizhi capsule and the western medicine group significantly increased.Cells in these groups were arranged closely and the cell structure was intact.3.The results of ELISA showed that compared to sham operation group,the VD group showed a higher serum level of AchE(P<0.01)and Bax level was significantly increased(P<0.01),and decreased serum level of Bcl-2(P<0.01).Compared with VD group,the serum level of AchE of the low-dose Sanshen Yizhi capsule group,the high-dose Sanshen Yizhi group and the western medicine group was significantly decreased(P<0.05),however the serum level of Bax was significantly increased(P<0.05).The serum level of Bcl-2 in the low-dose Sanshen Yizhi capsule group was not significantly changed,however significantly increased in high dose group and western medicine group was(P<0.01).Conclusion:1.Sanshen Yizhi capsule can improve the learning and memory ability in VD rats.2.Sanshen Yizhi capsule can improve the structure and morphology of nerve cells in hippocampus in VD rats and restore the morphological damage of hippocampus of VD rats.3.Sanshen Yizhi capsule can reduce serum AchE level,Bax level and increase Bcl-2 level in VD rats,it is suggested that Sanshen Yizhi Capsule can improve learning and memory ability and brain protection of VD rats by regulating hippocampal CA1 region.
