Objective: Intervention of heart-qi deficiency coronary heart disease in rats by Yiqi Huoxue prescription.The content of plasma of catecholamine(adrenaline and norepinephrine)and cAMP levels and the expression of Gs,PKA,AC,RyR2 mRNA in myocardial tissue are observed.Study on the protective effect and mechanism of Yiqi Huoxue prescription on coronary heart disease.Methods: The rat model of heart-qi deficiency of coronary heart disease is established by using the coronary artery ligation combined with intraperitoneal injection of the booster drug L-nitroarginine.Rats randomly divided into blank group,model group,western medicine group(diltiazem hydrochloride group),Yiqi Huoxue traditional Chinese medicine(TCM group is divided into low,medium,high concentration).After the successful replication of the model,intervention was given to the traditional Chinese medicine group and the western medicine group respectively.On the 45 th day of the experiment,The contents of plasma epinephrine(E),norepinephrine(NE)and cAMP and the expression of Gs,AC,PKA and RyR2 mRNA in rat cardiac tissues were detected.Result: The physical signs of the model rats are consistent with the symptoms of heart-qi deficiency of coronary heart disease.Through the detection and analysis of the plasma results and the comparison with the indexes in the early clinical trial stage,a similar trend was found: The results of plasma catecholamine and cAMP analysis showed that compared with the blank group,the model group increased significantly(p < 0.01),and compared with the model group,the high dose group of traditional Chinese medicine decreased significantly(P < 0.01).the expression of Gs,PKA,AC,RyR2 mRNA in the myocardium of rats are statistically significant.Compared with the blank group,the model group was significantly higher(p<0.01);Compared with the model group,high dose group of traditional Chinese medicine was significantly lower(p<0.01);There was a significant difference between the western medicine group and the high-dose group of traditional Chinese medicine(p < 0.01).Conlusion: Coronary artery ligation combined with intraperitoneal injection of L-nitroarginine is an ideal method for establishing rat model of heart-qi deficiency of coronary heart disease;Its effect on heart-qi deficiency of coronary heart disease may be mediated by regulating the contents of catecholamine and cAMP,mediating the Gs-AC-cAMP-PKA signal pathway to improve the functional abnormalities of RyR2.So it can protect the damaged myocardium.
