Objective:To investigate the effect of rat model of dampness on the oxidative metabolism of glucose in liver.Revealing the molecular mechanism of metabolic and energy metabolism imbalance in the cardiotoxic medium and the role of spleen and stomach in regulating glucose aerobic metabolism system and the intervention of Pingweisan.Method:Using the early mature method establish a animal model of dampness.Measured D-xylose ontent by Pyrogallol method,Na~+-K~+-ATPase activity was measured by chemical colorimetry.The content of glucose in serum was measured by chemical method.The protein、PK、PA of liver were detected by chemical method.The activity of Ac-coA、CS、SDH、RCCI、RCCIV were detected by ELISA.The activity of a-KGDHC、ICD in liver tissue was detected by microanalysis and the content of ATP and ADP were detected by HPLC.The contents of NAD~+and NADH and CA were detected by spectrophotometry.Results:1 Establish the middle Jiao model(1)Through the general observation,compared with the blank group,the model group significantly reduced body weight,diet,mental state(2)The gastric emptying was delayed in the model group,the content of serum D-xylose and the activity of Na~+-K~+-ATPase was decreased.(3)After treatment with Pingweisan,the body weight,mental state,gastric residual,intestinal propulsion,serum D-xylose absorption and Na~+-K~+-ATPas activity were improved in the model group.Damp-blocking model of rat model was successful.2The related factors of glucose metabolism in cytoplasm of liver by middle Jiao modelCompared with the blank group,the serum glucose content in the model group was significantly higher than that in the blank group,and the activity of pk and pc were increased in liver tissue.Compared with the model group,the serum glucose content in the natural recovery group was decreased,the activity of pk and the content of pc in the liver tissue continue to increase.The content of pk,pc in the liver tissue of the rats in the high dose group were higher than those in the blank group,and the change of the content of pyruvate in the rat liver tissue were higher than those in the blank group Consistent with the flat stomach scattered low dose of the most obvious.3The metabolic Stage of Glucose Metabolism in Liver effect by middle Jiao model3.1 The content of acetyl-coa effect by middle Jiao modelCompared with the blank group,the content of ac-coa in the liver tissue of the model group showed a decreasing trend.Compared with the model group,the content of ac-coa was still decreased in the natural recovery group,The content of acetyl-coa in the tissues was negatively correlated with the dose size,and the low dose group was close to the level of blank group.3.2 The Key Enzymes and Related Regulators in TAC effect by middle Jiao modelCompared with the blank group,the activity of CS,IDH,a-KGDHC,SDH and CA content were increased in the model group.Compared with the model group,the activity of CS and SDH in the natural recovery group from the liver tissue of were decreased,the IDH,a-KGDHC Enzyme activity and a-KGC content were increased,CA content changes do not have a downward trend.The activity of CS and a-KGDHC increased in the liver tissue of rats,and the activity of IDH,SDH and a-KGC were decreased,CA content tends to be stable.3.3 The Correlative Factors in Oxidative Phosphorylation effect by middle Jiao modelCompared with the blank group,the activity of rcc1,rcc4 and ATP,NAD~+and NADH were decreased in the liver tissue of model group,the change of ADP content was not stable.Compared with the model group,the activities of rcc1,rcc4 and the content of NAD~+withNADH were increased in the natural recovery group,the contents of ATP and ADP were decreased.After treatment,the activity of rcc1,rcc4and content of ATP,ADP,NAD~+and NADH were increased in rat liver tissue.Conclusion:The experimental results show that the model of humidification can affect some factors related to the oxidative metabolism of glucose in the liver,which may be one of the mechanisms of cell metabolism and energy metabolism imbalance in the dampness resistance.Pingweisi can improve the wet resistance model Liver glucose oxidative metabolism system partial factor defect status,involved in material metabolism and energy metabolism regulation of the regulation,which may be the regulation of glucose and glucose oxidative metabolism of the role of the target system and the stomach of the role of wet and spleen spleen one of the mechanism.
