Objective: To investigate the accumulation of dampness in middle Jiao syndrome animal model of synergistic effect of transporter expression in duodenal water-glucose synergistic transporter and the relationship with the intervention of Pingweisan powder.Revealing some of the mechanism of water metabolism in the dampness and Pingweisan control targets of duodenal fluid metabolism.Method: Adopting the early maturity of the dampness in middle Jiao syndrome animal model,by measuring the gastric residual rate and intestinal propulsion rate in rats,the content of D-xylose was determined by using the pyrogallol method,determination of Glucose in Serum by Peroxidase Method,the content of GLUT1,GLUT2,GLUT3 and SGLT1 in duodenal tissue were measured by enzyme-linked immunosorbent assay(ELISA)and the expression of GLUT1,GLUT2,GLUT3 and SGLT1 in duodenal tissue were detected by immunohistochemical SP method and mean optical density.Results:1 Syndrome of dampness in middle Jiao model:(1)Compared with the untreated rats,the body weight of the model group decreased significantly,the diet was decreased and the mental state was worse.(2)Objective indicators of detection,the model group had gastric emptying delay,bowel hyperactivity,serum glucose increased,serum D-xylose content decreased.(3)After treatment with Pingweisan powder,in the model group,the body weight,mental state,gastric residual rate,intestinal propulsion rate,serum glucose and serum D-xylose absorption were improved.It is suggested that the rat model of dampness syndrome is successful.2 The content of water-glucose synergistic transporter in duodenal Compared with the normal group,the content of GLUT1 and GLUT2 in the duodenum of the rats in the model group were significantly higher than those in the control group,while the GLUT3,SGLT1 content decreased or decreased the trend.Compared with the model group,The content of GLUT1,GLUT2 and GLUT3 in the high dose and middle dose groups decreased decreased the trend.The content of GLUT1 and GLUT3 in the low dose group were increased,while the content of SGLT1 in the low dose group was significantly decreased.After drug treatment,compared with the rats in the natural recovery group,the content of GLUT1 in the middle dose and low dose group were increased,the content of GLUT2 in the high dose and low dose group were increased,the content of GLUT2 in the middle dose group decreased,the content of GLUT3 in the high and low dose groups were decreased.The content of SGLT1 in the high、middle and low dose groups were decreased.Rats were given flat stomach powder,compared with the normal group,the content of GLUT1 in rats was higher than that in blank group,and the content of GLUT3 in rats decreased.3 Expression distribution of water-glyco-synergistic transport protein in duodenum Compared with the normal group,GLUT1,GLUT2,GLUT3 and SGLT1 were upregulated in the duodenum of the model group.Compared with the model group,the expression levels of GLUT1,GLUT2 and GLUT3 were down-regulated in the high and middle dose groups and the expression of SGLT1 in the high、middle and low dose groups was down-regulated.After drug treatment,compared with the natural recovery group,the expression levels of GLUT1 and GLUT3 were down-regulated in the high dose and middle dose group,and the expression levels of GLUT2 and SGLT1 were down-regulated in the middle and low dose groups.Conclusion: The ocontents of GLUT1 and GLUT2 in the duodenum of rats exposed to the model were significantly increased,while the contents of GLUT3 and SGLT1 decreased or decreased,GLUT1,GLUT2,GLUT3,SGLT1 were upregulated in the duodenal expression level,this may be caused by dampness in the cesarean syndrome in the small intestine to clear the secret disorders,water infusion intestinal intestinal lax diarrhea caused by one of the mechanisms;Pingweisan by reducing the GLUT1,GLUT2,GLUT3,SGLT1 content changes and downgrade GLUT1,GLUT2,GLUT3,SGLT1 expression levels,mediators of duodenal aqueous fluid abnormalities;Normal rats were given Pingwei powder,compared with the normal group,the content of rat GLUT1 increased,the content of GLUT3 in rats decreased,and the GLUT1、GLUT2、GLUT3、SGLT1 expression levels were raised,suggesting that flat stomach scattered in case of non-dialectical use of water metabolism caused by one of the mechanisms of imbalance.
