| Objective To explore the role of specific regulation of Baihui acupoints in different functional states of the body,so as to elucidate the specific expression of meridian points in Baihui acupoints.Methods Male Sprague-Dawley rats weighing 200-250 g in Experiments 1 and 2 were randomly divided into normal blank group,normal Baihui group,model blank group,and model Baihui group.The model group was prepared epilepsy rat model,the normal group was injected with the same volume of saline.The interventions given to the model blank group and the normal blank group were only fixed,not acupuncture,once a day for 30 minutes;the model Baihui group and normal Baihui group were fixed and acupuncture Baihui,1 each time,30 min;10 days later,the rats of each group were taken for treatment and the corresponding indicators were observed.Experiment I: The behavioral changes of epileptic rats during acupuncture were monitored by monitoring equipment for 24 hours,and the expression of immunocytic factors in hippocampal tissue of rats in each group was detected by liquid phase microarray technique.Experiment 2: The histopathologic changes of hippocampal tissue were observed by HE staining and the apoptosis rate of hippocampal neurons in each group was detected by flow cytometry.Results Experiment 1: 1.Behavioral changes:(1)Frequency of epileptic seizures in rats:On the first day of acupuncture,the number of epileptic seizures in the model Baihui group was slightly higherthan that of the model blank group,and the seizures increased more and more frequently.After the fifth day,the overall frequency of episodes in the model Baihui group was lower than that in the model blank group.On the seventh day,the number of seizures in the model Baihui group showed a significant declining trend,and the rats in the model blank group continued to have frequent seizures.(2)The duration of epileptic seizure in rats: On the first day of acupuncture,there was no significant difference in seizures between the two groups.From the next day,the seizure duration of the model blank group rapidly decreased until the fifth day,while the model Baihui group showed a slow decline.After the fifth day,the model blank group showed a fluctuating upward trend,and the overall duration was longer than that of the model Baihui group.However,the attack duration of the model Baihui group was not changed on the 5th to the 8th day,and there was a significant downward trend after the eighth day,both are lower than the model blank group.(3)Grades of seizures in rats: On the one hand,rat III epileptic seizures last longer,followed by grade IV,and grade V seizures have a relatively short duration.After acupuncturing Bahui,the duration of each episode was generally decreasing.On the other hand,seizures in rats mainly occurred in grade IV,followed by grade III,with the least number of episodes in grade V.After acupuncturing Bahui,the total number of seizures was significantly reduced.2.Cytokine expression:Compared with the normal blank group,the expression levels of six cytokines such as Ig M,Ig G1,Ig G2 a,Ig G2 b,Ig G2 c,and Ig A in the model blank group showed a significant upward trend,among which Ig M,Ig G1,Ig G2 a,Ig G2 b,and Ig A were significantly different(P<0.05);the normal blank group compared with the normal Baihui group,six cytokines were not statistically significant(P>0.05);Compared with the model blank group,the expression levels of Ig M,Ig G1,Ig G2 a,Ig G2 b,Ig G2 c,and Ig A cytokines in the model Baihui group decreased to varying degrees,and the expression levels of Ig G2 a and Ig A were statistically significant(P<0.05);Model Baihui group compared with the normal Baihui group,Ig G2 a,Ig A expression was statistically significant(P<0.05).Experiment 2: 1.HE staining results:(1)In rat hippocampal CA1 area,the number of positive cells in normal Baihui group was lower than that in normal blank group,and that in model Baihui group was lower than that in model blank group(P>0.05);the number of positive cells in model blank group was significantly higher than that in normal blank group,the model Baihui group was higher than the normal Baihui group(P<0.05).(2)In rat hippocampal CA3 area,the model blank group was higher than the normal blank group and the model Baihui group was higher than the normal Baihui group(P<0.05);after acupuncturing Baihui,the number of positive cells in both normal and epileptic rats decreased(P<0.05).(3)The rat hippocampal DG area,model blank group was higher than the normal blank group,the model Baihui group was higher than the normal Baihui group(P<0.05);compared with the normal blank group,the number of positive cells in the normal Baihui group decreased significantly(P<0.05);compared with the model blank group,the model Baihui group also significantly reduced(P<0.05)2.Results of neuronal apoptosis rate:(1)Early apoptotic conditions:The apoptosis rate of nerve cells in the normal Baihui group was higher than that of the normal blank group,but it was lower than that of the model Baihui group.The model blank group was slightly higher than that of the model Baihui group(P>0.05);The apoptosis rate of the blank group was significantly higher than that of the normal blank group(P<0.05).(2)Late cell apoptosis: Compared with the normal blank group,the apoptosis rate of the normal Baihui group was increased(P>0.05);the model Baihui group was significantly higher than the model blank group and the normal Baihui group,and the model blank group was higher than the normal blank group(P<0.05).Conclusion 1.Acupuncture at Baihui has specific bidirectional effects on the morphological changes of neurons in CA3 and DG regions of rat hippocampus under normal and epileptic conditions,while the regulation of the apoptosis of immunocytes Ig A,Ig G2 a and hippocampal neurons was specific only in the epileptic state of rats.2.The mechanism of acupuncture at Baihui points on epilepsy may be related to the decrease of the expression of immune factors Ig A and Ig G2 a in hippocampus. |
