Objective: To explore the mechanism of Fuzheng Tongdu on the hippocampal protection in epilepsy rats,and then to explain the specificity of meridian effect in different body condition and multi-level target comprehensive regulation.Methods: 72 male SD rats were randomly divided into Model group(42)and Normal group(30),and the Model group was divided into 3 groups: Model blank group(group AM),Model control group(group BM),Model meridian group(group CM),the Normal group was divided into 3 groups: Normal blank group(group AN),Normal control group(group BN),Normal meridian group(group CN).The model group was seted up epilepsy model though injecting lithium chloride-horse roca while normal group was injected with saline solution.The treat plan is: AM group and AN group was fixed 1 time a day,every time 30 min,but do not acupuncture.BM group and BN group acupuncture 1 time a day,every time 30 min,acupoint including Waiguan,Shousanli and Huantiao,while BM group and BN group acupuncture 1 time a day,every time 30 min,acupoint including Dazhui,Baihui and Housanli,rats in each group were treated with hippocampus for 10 days.Observing the rats weight and mortality,rats hippocampal tissue pathological morphological changes was tested by He staining.The hippocampal neuron apoptosis rate was detected by flow cytometry.Result: 1.The mortality of CM group was decreased compared with AM group,The epileptic rats lost weight compared with normal rats,(P<0.05).2.Histopathology of hippocampus:(1)The positive cell rate of CA1 and CA3 in hippocampus of AM group was significantly higher than that in AN group,and the positive cell rate in DG area was significantly lower.(P<0.05).(2)The positive rate of CA1,DG and CA3 in hippocampus of CM group and BM group were significantly lower than those in AM group,and the positive rate of CA1 area in CM group was lower than that in BM group,(P<0.05).(3)The positive rate of CA1,DG and CA3 in hippocampus of CN group was lower than that in AN group.The positive rate of CA1 and DG in BN group was lower than that in AN group,and the positive rate of DG3 and CA3 in CN group was lower than that in AN group(P<0.05).(P<0.05).(4)In the three hippocampus of the rat hippocampus,the positive cell rate and the positive cell ratio of CA1 in the AN group were the lowest,and the positive cell rate and the positive cell ratio in the CA1 region of the AM group were the highest.3.Immune rate of hippocampal neurons in rats:(1)The apoptotic rate of early and late hippocampal neurons in AM group was higher than that in AN group(P<0.05).(2)There was no significant difference in the early hippocampal neurons between the Model groups(P> 0.05),but the CM group and the BM group were lower than those in the AM group,and the BM group was the lowest.There was no significant difference in the late hippocampal neurons between the Normal groups(P> 0.05),but the CM group and the BM group were higher than those in the AM group,and the CM group was the highest(P<0.05).(3)In the Normal group,The early apoptotic rates of CN group and BN group were higher than those of AN group(P<0.05),and the BM group was the highest(P<0.05);the late apoptotic rate of the CN group and the BN group were higher than that in AN group,and the CN group was the highest(P<0.05).Conclusion: 1.Fuzheng Tongdu can reducing the mortality of epilepsy rats,the mechanism may be related to the reduction of rat hippocampal neuron positive cell rate.2.The specificity of the acupoint effect has obvious targeting in hippocampal histomorphology and neuronal apoptosis,and relate to the different body condition.
