| Objective:Observe the effect of acupuncture Baihui acupoint on vascular regeneration related factors in cerebral ischemia-reperfusion rats,to explore the mechanism of acupuncture Baihui point in the treatment of cerebral infarction,and to provide theoretical basis for acupuncture Baihui point in the treatment of cerebral infarction.Method:A total of 72 healthy male SD rats were selected,By using the method of random number table,it was divided into 3 groups,sham operation group,model group,acupuncture group,24 in each group,and then randomly divided into 3 subgroups according to the time point 1d,3d,7d,each subgroup 8.Keep in cage according to group,eat and drink freely,keep 12 hours for day and night change.To prepare the model of cerebral ischemia reperfusion in middle cerebral artery.The sham operation group and the model group did not give the treatment,only carried on the same condition grasping.Acupuncture group was given acupuncture Baihui point treatment.In 1d,3d,7d,The modified neurological function defect score of rats was observed,the local cerebral blood flow was measured by Doppler ultrasound,the volume of cerebral infarction was observed by TTC staining,the morphology of brain tissue was observed by HE staining,and the changes of vascular related factor VEGFR-2,Ang-1,Tie-2 were observed by immunohistochemistry.Result:1.The behavioral changes of rats were as follows: there were no symptoms of nerve function defect in the sham operation group,and the symptoms of nerve function defect appeared in the other groups.The scores of acupuncture group 3d,7d,after cerebral ischemia were lower than those in the model group(P<0.05).2.Cerebral blood flow measurement: brain blood flow chart of sham operation group is normal.The cerebral blood flow in the model group and the acupuncture group decreased,and the cerebral blood flow in the acupuncture group and the model group increased at any time,and the cerebral blood flow in 3d,7d acupuncture group was higher than that in the model group(P<0.05).3.Cerebral infarction volume: no cerebral infarction in sham operation group.The infarct area appeared in both model group and acupuncture group,and the volume of cerebral infarction decreased in 1d,3d,7d,model group and acupuncture group,and the volume of cerebral infarction in acupuncture group was lower than that in model group at 3d,7d(P<0.05).4.Changes of brain tissue morphology: no changes in brain tissue morphology,neat arrangement of brain cells,full cerebral vessels.All the time points of model group and acupuncture group showed obvious changes of blood samples in brain tissue.At each time point,the brain tissue morphology of acupuncture group was better than that of model group,and the curative effect d acupuncture 7d was the best.5.The changes of VEGFR-2,Ang-1,Tie-2: the positive expression of protein in each group except sham operation group increased in different degrees at1 d,3d,7d time point,and the positive expression of each time point in acupunct ure group was higher than that in model group(P<0.05).Conclusion:1.Acupuncture at Baihui acupoint can reduce the nerve function defect score in cerebral ischemia-reperfusion rats,increase the brain blood flow,reduce the volume of infarction,and promote the repair of pathological tissue in rats with cerebral infarction.2.Acupuncture at Baihui acupoint can promote the increased expression of VEGF R-2,Ang-1 and Tie-2 proteins in the brain tissue of cerebral ischemia and reperfusion,promote microangiogenesis and improve the microcirculation of brain tissue in the ischemic area.3.Acupuncture at Baihui acupoint in the treatment of cerebral infarction may be related to the expression of VEGFR-2,Ang-1,Tie-2 protein. |
