Study On Tissue Culture Of Veronica Rotunda

This paper explored tissue culture technology of Veronica rotunda's rapid propagation,and used to provide theoretical basis and experimental evidence for efficient propagation of the wild flower resources.The sterile plants were studied by using stem with axillary buds as explants,callus induction,adventitious buds technology,proliferation culture,rooting andtransplanting acclimation series of technical researches were conducted.The main results as following:1.Leaves with the most appropriate method of disinfection about V.rotunda were explanted as 75%alcohol,15 s and 10%sodium hypochlorite,10 minutes.The contamnat-ion rate was 10.00%,the browning rate was 33.33%,the survival rate was 56.57%.2.The best explant for inducing auxiliary buds of V.rotunda was MS+6-BA1.0 mg/L+NAA0.3 mg/L+ GA₃1.0 mg/L,then the inductivity was 83.33%,the inducible coefficient was 1.41.3.Leaves was the best explant for inducting callus,putting the leaves upwards,the best induction culture medium was MS+2,4-D0.2 mg/L+NAA0.05 mg/L+6-BA0.5 mg/L,then the callus induction rate was 100.00%,and the callus was green,loose texture,without browning.4.The best culture medium for inducing callus to differentiate into adventitious buds was MS+2,4-D0.1 mg/L+6-BA1.0 mg/L +NAA0.1 mg/L,then the differentiation rate was 31.11%.5.The best multiplication medium of V.rotunda was MS+6-BA1.0 mg/L+NAA0.1 mg/L,and the multiplication coefficient was 2.87,the average height was 5.0 cm,and seedlings grown robust.6.The best rooting medium of V.rotunda was 1/2MS+IBA0.5mg/L+NAA0.3mg/L,and the rooting rate was 96.67%,the average number of roots was 6.85,the average root length was 4.67 cm,and roots grown in good condition.7.The transplanting domestication method of V.rotunda was that,under the natural light irradiation with opening bottle domestication 3 days,planting seedlings in vermiculite:perlite:peat?1:1:1?matrix.
