The stems of P. tomentosa clone-30, he stems and leaves were used as the explants to study their regeneration and regeneration conditions, in order to build their rapid tissue propagation system. The results of the experiment show that:1. Considering the explants lowest rate of browning and the lowest rate of pollution, to the both of the material, the most suitable disinfection method was84disinfectant3min and0.1%of the Hgcl24.5min. The most suitable basic medium was MS medium.2. Through the induction of the use of cytokinin to the P. tomentosa clone-30, we found that, when usd0.5mg/L6-BA to induce the axillary buds, the highest induction rate was86.7%.3. The Orthogonal test showed that:NAA and6-BA have the most important influences to the buds proliferation of stems, different kinds of medium had no significant influences. The most suitable medium when used6-BA/NAA to induce the stem segments of P. tomentosa clone-30was MS+0.7mg/L6-BA+0.05mg/LNAA, the proliferation of the adventitious buds was628%.4. For the P. tomentosa clone-30, the most appropriate rooting medium was1/2MS+NAA0.01mg/L, the rooting rate was96.8%,the average number of rooting was6.3, the average root length was4.6cm. The colour of the plant was bright green,the root system was robust and the plants growed nicely.5. The upper stem was the appropriate explant for the hybrid of white populus02-9-22, the induction rate of adventitious buds was86.3%.6. The test of cytokinin inducted adventitious buds showed that:the appropriate concentration of6-BA to induct the stems adventitious buds of02-9-22was0.5mg/L, the induction rate of stems adventitious buds was80.0%; the appropriate concentration of6-BA to induct the leaves cluster buds of02-9-22was0.7mg/L, the induction rate of leaves cluster buds was83.3%.7. The orthogonal test showed that:6-BA and NAA have the most important influences to the buds proliferation of stems about02-9-22, different kinds of medium had no significant influences. The most suitable medium when used6-BA/NAA to induce the stem segments of02-9-22was MS+0.3mg/L6-BA+0.15mg/LNAA, the proliferation of the adventitious buds was574%. The most suitable medium when used6-BA/NAA to induce the leaves cluster cluster of02-9-22was1/2MS+0.3mg/L6-BA+0.10mg/LNAA, the proliferation of the cluster buds was484%.8. For the hybrid of white populus02-9-22, the most appropriate rooting medium was1/2MS+NAA0.05mg/L, the rooting rate was93.3%,the average number of rooting was6.4, the average root length was4.8cm. The plant was height, the blade was big and dark green, the plant growed strong and the root was highest developed.
