Acer×freemanii stem with axillary bud was used as explant,tissue culture of Acer×freemanii was studied systematically in the article in order to set up the technique for commercial production of Acer×freemanii and provide technical support for molecular breeding and correlation study of Acer×freemanii.During the establishment of tissue culture technique system, Acer×freemanii'Marmo','Autumn Blaze'stem with axillary bud was used as explant. In this study,the nodal position, collection time, the degerming results using different germicidal treatment programs, the control of browning method, the optimal initiation medium,the optimal proliferation medium, the optimal rooting medium in Acer×freemanii's tissue culture were studied systematically. The results indicated that:1) The optimal collection time was the last ten days of April to May.2) The optimal nodal position was the third and the forth nodes of the new-born stem.3) Drawing the explant in May, the optimal germicidal treatment program was: 75% C2H5OH 20s+0.1% Hgcl2 4min.4) The optimal initiation medium in tissue culture of'Marmo'and'Autumn Blaze'was MS.5) The browning degree in'Marmo'and'Autumn Blaze'was similar, they were"V"type change in the browning degree with the different bud location and all the peak amplitude was the fourth bud. The optimal browning inhibitor about'Marmo'was PVP1.0mg/L,and the optimal browning inhibitor about'Autumn Blaze'was PVP 0.5 mg/L.6) There were different effect tendency of plant growth regulating substance on the proliferation of different Acer×freemanii plantlets,but TDZ was the dominating effect factor to proliferation of plantlets, NAA was not the dominating effect factor.The optimal proliferation medium of'Marmo'was MS+TDZ 0.02mg/L ,the optimal proliferation medium of'Autumn Blaze'was MS+TDZ 0.05mg/L.7) The effect of NAA on rootage of different Acer×freemanii plantlets was significant, the effect of AC was very important too. The optimal rooting medium of'Marmo'was MS+NAA 0.1 mg/L+AC 0.5g/L, The optimal rooting medium of'Autumn Blaze'was MS+NAA 0.2 mg/L+AC 0.6g/L.
