Panax ginseng and Panax quinquefolium belong to Araliaceae Panax plants and have a variety of physiological and pharmacological functions,of which ginsenoside is the main active component and secondary metabolites. But the low content, especially for the rare ginsenoside, serious influenced its’ development and application. How to improve the yield and ginsenoside content has become a main research topic. The synthesis and accumulation of plant secondary metabolites has closely related with plants defense responses. The synthesis is highly regulated by the plant body itself. Plant defense response will be activated only after external infringed and start for the secondary metabolites biosynthesis. The plant defense response was caused by many factors including microbial infection, endogenous or exogenous gene, soil, temperature, light etc. Paenibacillus polymyxa has the function of transforming ginsenosides in vitro, cinder can be pervious to water and Ventilationcan which can improve the function of the soil, but whether can they promote the accumulation of ginseng plant growth and secondary metabolites of ginsenoside still need experiment to prove. The mechanism of Rolc gene of Ri carrier which can promote the accumulation of ginsenoside Rb1 needs to be further studied. Therefore, this experiment carried out 2 Influence Factors(P. polymyxa and cinder) on the study of Panax ginseng, Panax quinquefolium growth and ginsenosides accumulation. Yeast two hybrid technique was used to screen the proteins interacting with rolc gene in ginseng. Thus, the study offers the support and reference for regulation of ginsenoside biosyntheses. The results are as follows:1. P. polymyxa is an endophytic bacteria isolated from ginseng. It can promote growth and ginsenosides accumulation of 1- to 4-year-old ginseng and reduce the incidence of ginseng growth period(P<0.05) when applied it by foliar application, root irrigation or combined treatment of the two. The effect of foliar application and root irrigation combined on treatment group was significantly better than the foliar application treatment group, irrigation treatment group and the control group(P<0.05). The height of 1 to 4 years old ginseng increased 38.44%,35.24%,41.90% and 24.00%, respectively; The fresh weight increased 31.64%,58.87%,46.70% and 18.61%, respectively; The morbidity decreased 13.64%,17.67%,21.67% and 27.34%, respectively; Rt(the sum of 9 gisenosides) increased 36.83%,44.52%,67.96% and 79.44% respectively.2.Ginseng field which was modified by cinder can improve Panax ginseng and Panax quinquefolium germination rate and seedling rate, enhance seedling growth and 9 kinds of ginsenosides accumulation of Panax ginseng and Panax quinquefolium(P<0.05). The optimal size of cinder modification of ginseng land is 20 mesh, the best ratio is 6kg/m2.When the size of cinder is land 20 mesh, the germination rate and seedling rate were 68.11% and 78.25% of Panax ginseng; The germination rate and seedling rate are 71.78% and 82.10% of Panax quinquefolium;Rt which stands for the sum of 9 ginsenosides inceased 44.41%(Panax ginseng) and 32.78%(Panax quinquefolium). When the best ratio of 6kg/m2, ginseng germination rate and seedling rate were 64.22% and 75.09%;The Panax quinquefolium germination rate and seedling rate were 70.33% and 79.11%;Rt inceased 24.21%(Panax ginseng) and 28.02%(Panax quinquefolium).3. Completed construction of PGBKT7-rolc bait rolc gene recombinant vectors and 2 new ginseng protein genes interacting with rolc gene could promote the growth and accumulation of ginsenosides in ginseng had been screened.
