Objective:The objective of this research is to study and analyze the effect of Yiqi YangyinGuchong prescription and its disassembled prescriptions on the weight, uterus index,hemorheology, endometrial pathomorphology, endometrial aquaporin1(AQP1) andthrombospondin-1(TSP-1) of anovulatory dysfunctional uterine bleeding model rats withQi-Yin deficiency syndrome by performing animal experiment. The composition principles ofYiqi Yangyin Guchong prescription would be investigated in order to lay a foundation forfurther optimization, and to provide an experimental basis for its further study and clinicalapplication.Methods:70healthy SD female rats of SPF level were chosen, and randomly separated a groupfrom them and would be regarded as the blank control group in which rats was performedsham-operation, normally fed and saline filled daily by gavage. Meanwhile, the other ratswere castrated with bilateral ovary excision and impelled to swim in the water tank understarvation and fatigue for14days after wound healing, filled with progynova solution bygavage at the same time, and hydrocortisone injection for4days since the11th day. On the15th day, rats in the model group were divided randomly into model control group,Gongxuening group, Yiqi group, Guchong group, Yangyin group and omni-prescription groupon the very next day, which, except the model control group, filled with correspondingmedicine by gavage for5days. Rats in the model control group and the blank control group,however, were filled with saline daily at the same time. All the rats would be extirpated40minutes later after the last gavage to weigh the uterus and test hemorheology, endometrialpathomorphology, AQP1and TSP-1.Results:1.Compared with the blank control group, rats in each model group have a deficiency ofboth Qi and Yin. During post-medication period, the general condition of rats in medicationgroups were obviously improved, but with no significant difference.2.Compared with the blank control group, the physical weight of the model groups ratsand post-medication groups rats respectively decreased and increased significantly, whosedifference(P<0.05)was of statistical significance; Compared with the blank control group,the uterus index of the model groups rats and post-medication groups rats respectivelyincreased and decreased significantly, whose difference (P<0.05) was of statistical significance; while the difference among post-medication groups was of no statisticalsignificance(P>0.05).3.Compared with the blank control group, rats in each model group appeared obviousendometrial thickening, disordered arrangement of glandular epithelium cells, glandincreasing and glandular cavity extending. These symptoms were improved significantlyduring post-medication period, especially in omni-prescription group and Yiqi group.4.Compared with the blank control group, hemorheology related indexes of rats in allmodel groups increased significantly (P<0.05); while compared with the model controlgroup, these indexes in all post-medication groups declined significantly and were ofstatistical significance (P<0.05), but no statistically significant difference appeared amongthe medication groups (P>0.05).5.Compared with the blank control group, OD value and MVD (which is positive stained)of AQP1and TSP-1’s in all model groups respectively decreased and increased significantly,which two difference were both of statistical significance (P<0.05); moreover, anegative correlation existed between AQP1and TSP-1, which was of statistical significance(r=﹣0.963, P<0.01). The above expression would be improved significantly duringpost-medication period (P<0.05), especially in Yiqi group and omni-prescription group (P<0.05), which two, however, has no significant difference (P>0.05); the difference amongother medication groups was also of no significance (P>0.05).Conclusion:1.The Yiqi Yangyin Guchong prescription can improve the general condition (like weight,uterine wet weight, uterus index,etc.) as well as endometrial morphology of anovulatorydysfunctional uterine bleeding rats with Qi-Yin deficiency syndrome. Among its disassembledprescriptions, the medicine in Yiqi group and omni-prescription group can invigorate qi andstanch significantly by enhancing endometrial self-recovering function.2.The Yiqi Yangyin Guchong prescription can regulate fluidity and glutinousness inblood, improve the dense and glutinous blood condition, and relieve endometrial hemorrhageand yin deficiency symptoms by invigorating qi, invigorating pi, yang yin and gu chong.3.The Yiqi Yangyin Guchong prescription can increase AQP1’s protein expression andits MVD (which is positive stained) but reduce TSP-1’s protein expression and its MVD in theendometrium of anovulatory dysfunctional uterine bleeding rats with Qi-Yin deficiencysyndrome. This prescription could achieve therapeutical effect on anovulatory dysfunctionaluterine bleeding with Qi-Yin deficiency syndrome by improving angiogenesis of endometrial microvessel and endometrial self-recovering function. |
