Objective:To esttblish a spontaneity hypertension rat model with liver prosperity and phlegm obstruction syndrome, and observe the effect of yun ke ping granule on blood pressure, blood fat and angiotensin Ⅱ of spontaneity hypertension rat with liver prosperity and phlegm obstruction syndrome, meanwhile explore its possible underlying mechanismsMethod:Fourty SHR were randomly divided into low or high does of yun ke ping granule, suspension of Captopril and normal saline groups, all of them were established the liver prosperity and phlegm obstruction tsyndrome by a slow irritation and fat-rich diet. By observing the dynamics of the temperament and weight, blood pressure, blood fat and angiotensin Ⅱ to confirm to have established the model of SHR with liver prosperity and phlegm obstruction syndrome. Then each group was respecti vely drenched with low or high does of yun ke ping granule, the suspension of Captopril, and normal saline, all these groups were drenched for consecutive thirty days. After the first intragastric administration, the tail artery systolic pressure of the rats was measured, meanwhile the changes of blood fat and angiotensin Ⅱ were determined at the end of the8th week.Result:Animal behavior observation results:①The performance of model group is agitated, irritable, at the same time they fought each other, the color of eye conjunctiva deepened and turned red; Rotation time measure:The Rotation time of the model group shortened significantly, and had a significant difference compared with controls (P<0.05).②In the hypotensive experiment, low or high dose of yun ke ping granule and Captopril could reduce the tail artery systolic pressure (P<0.05), while the effect of high does of yun ke ping granule was better than that of low does (P<0.01), and the effect of Captopril was better than that of high does and low does (P<0.01).③Blood biochemical test results:Angiotensin Ⅱ in the group of low or high does of yun ke ping granule and Captopril significantly decreased (P<0.05), while the change of Captopril had no significant difference compared with that of high does and low does (P>0.05).④TG and TC had decreased significantly in both the yun ke ping groups(P<0.05), but there wasn’t a significant change in the Captopril(P>0.05).Conclusion:①The model of spontaneity hypertension rat with liver prosperity and phlegm obstruction syndrome could be established by a slow irritation and fat-rich diet.②Yun ke ping granule could reduce the blood pressure and blood fat and Angiotensin Ⅱ of SHR with liver prosperity and phlegm obstruction syndrome. |
