Objective:(1) Explore the method of intervention in rats with sick sinus syndrome model, to establish a small, simple and accurate modeling method with small damage. (2) To find the most suitable drug for the establishment of SSS model in formaldehyde, sodium hydroxide, acetic acid, ethanol, oil of Brucea javanica. (3) To summed up experimental methods in preparation for further study by comparing the mortality and the cause of death in each group.Methods:Select 60 rats with heart rate between 350 and 450 times/ min from healthy rats, and the 60 rats were randomly divided into six groups (10 in each group), formaldehyde routine group, formaldehyde intervention group, sodium hydroxide intervention group, glacial acetic acid intervention group, alcohol intervention, javanica oil intervention group. Formaldehyde routine group:use urethane for anesthesia, and connect breathing machine, then expose sinus node and wet compress sinus node area with 40% formaldehyde in a small gauze piece; formaldehyde intervention group:use anhydrous ether for anesthesia, and cut a small hole at the right edge of the second rat intercostal chest sternum, and insert the self-made mapping electrode along the mouth of this small chest in accordance with the ECG waveform to determine the administration site,and then drip 40% formaldehyde solution with a micro-syringe Mapping Catheter.Other intervention group use the same experimental methods with formaldehyde intervention group.Measure HR, electrophysiological, pathological observation, postoperative survival and the success rate of modeling to do the comparative study.Results:(1) Mortality rates of the routine group were higher than the intervention group, while there was no significant difference among the five intervention groups. (2) Heart rate of six groups all decreased,and rat heart rate of the formaldehyde routine group, formaldehyde intervention group, sodium hydroxide intervention group, glacial acetic acid intervention group decreased by 30% which meets the standards for model,while that of alcohol intervention and javanica oil intervention group decreased no more than 30% which does not meet the standards for model. By comparison, sodium hydroxide intervention group has the highest success rate of modeling.Conclusions:(1) Intervention modeling approach simplifies the modeling method of thoracic and improve the success rate of modeling. (2) Formaldehyde, sodium hydroxide can be used to establish the SSS model.
