Objective In this experimental study,we used the sodium hydroxide(NaOH)intervention method to create a rat sinus model,and on this basis,using traditional Chinese compound FuXinMai to intervene and observe its sinus rat model.The changes of electrophysiological function of atrial node were further explored to further explore the mechanism of the effect of FuXinMai on the electrophysiological function of sinus node in sick sinus model rats.Methods1.A rat sinus model was established.Rats were anesthetized and connected to a motor.A longitudinal incision of approximately 0.1 cm was made at the second intercostal surface of the right sternal border.A self-made electrode catheter was inserted along the opening into the chest and the VI electrocardiogram was plotted.The waveform is positioned at the administration site,and a 10%sodium hydroxide solution is microinjected with the self-made mapping electrode catheter.When the heart rate drops by 30%-50%,the injection is stopped.After continuous monitoring for 2 hours,the model is successfully established when the heart rate no longer fluctuates.The heart rate was measured before and 2h after modeling to determine the model effect.2.Three days after the model was established,after the rat model was stable,the Chinese compound FuXinMai was used to intervene the sick sinus rat model.The following indicators were monitored and recorded:(1)AA interval(PP interval);(3)Sinus node recovery time(SNRT)and corrected sinus node recovery time(CSNRT);(3)Sinus atrial conduction time(SACT).To further explore the effect of FuXinMai on the sinoatrial node electrophysiological function of the sinus node model rats.Results1.The success rate of establishing a sick sinus syndrome disease model in rats with NaOH intervention was high(90%),which overcomes the shortcomings of previous animal models such as susceptibility to infection,excessive bleeding,and need for ventilator support.Furthermore,the rat sinus model established by NaOH intervention method has the following advantages:(1)The diseased sinus animal model has high stability;(2)Long postoperative survival time can be used for time-consuming drug efficacy experimental study;(3)Postoperative infection rate is low.2.Compared with the control group,the high and middle doses of FuXinMai can effectively reduce the AA interval,SNRT,CSNRT,and SACT in the diseased sinus rat model,and the difference was statistically significant(P<0.01).Compared with groups in each dose group,the above values were significantly shortened in the high dose group compared with the middle and low dose groups,and the difference was statistically significant(P<0.05).Conclusion1.Sodium hydroxide intervention method for the preparation of sick sinus syndrome rat model has the advantages of accurate positioning,high success rate and low mortality rate.2.FuXinMai improve the electrophysiological function of sinoatrial node in sick sinus model rats,that is,it can shorten the AA interval,SACT,SNRT,CSNRT value of the sick sinus rat model in a dose-dependent manner.
