| Objective:To investigate the expression of Interleukin-1β(IL-1β) in STZ(streptozotocin)-induced diabetic retina,and to observe the effect of compound thrombolysis on IL-1βexpression.Method:52 Wistar rats were randomly divided into normal control group(N),diabetic group(D),low dose compound thrombolysis group (Câ… ),high dose compound thrombolysis group(Câ…¡).Group D,Câ… , Câ…¡were treated with intraperitoneal injection of 1%STZ(65mg/kg), aiming to establish diabetic rat model;Group N were treated with the same dose of sodium citrate - citric acid buffer intraperitoneal injection. Tail vein blood glucose was measured by the fast blood glucose instrument 7 days later.When blood glucose higher than 16.7mmol/ L,the rat was internalized into the diabetic model.Group Câ… were treated with 900mg/(kg·d) Compound Thrombolysis for intragastric administration,while group Câ…¡were treated with 1800mg /(kg·d) compound thrombolysis for intragastric administration,and group D were administered the same dose saline.The rats were killed at two weeks and four weeks.We investigate the IL-1βexpression level in each group by real-time PCR.Results:1.The blood glucose of Successful rat model was 20.23±2.58,and success rate reached 94.4%(34/36).The weight of successful rat model had no difference with control rats in 1 week(p>0.05),but had decreased statistical significantly after 2 weeks compared with control rats(p<0.01); The weight of rats in Compound Thrombolysis groups had no disparity compared with diabetic rats(p>0.05).2.Rather than the control group,the expression of IL-1βincreased 3.03~20.25 times in diabetic group,with significant statistical significance(P<0.01);The expression of IL-1βin diabetes group was higher on 4 week than on 2 week(P<0.05).3.The expression of IL-1βin Compound Thrombolysis intervention groups reduced statistical significantly(P<0.01) than in diabetic group; The expression of IL-1βin 900mg/(kg·d) dose Compound Thrombolysis groups(Câ… )had no difference with Group D(P>0.05) on 2 week,but 1800mg/(kg·d) dose group(Câ…¡) decreased statistically;The expression of IL-1βin Group Câ… and Group Câ…¡on 4 week decreased statistically (P<0.05) on 4 week than in Group D,and Group Câ… and Group Câ…¡had no difference(P>0.05).4.Rather than the normal control group,the expression of IL-1βin rat retina increased 3.61~6.02 times in group Câ… on 2 week,with statistical significance(P<0.05);The expression of IL-1βin rat retina in group Câ… on 4 week and in group Câ…¡on 2 week,4 week had no difference with in normal control group(P>0.05). Conclusion:1.The successful establishment of STZ-induced typeâ… diabetic rat model.2.The expression of IL-1βrised in diabetic rats' retina,and gradually increased with the time in 4 weeks.3.Compound Thrombolysis could inhibit the expression of IL-1β, making the IL-1βin diabetic rats' retina reaching the normal level; 900mg /(kg·d) dose and 1800mg /(kg·d) dose of Compound Thrombolysis had the same inhibition effectiveness,but 1800mg /(kg·d) dose reacted earlier than 900mg /(kg·d) dose. |
