According to duration of diabetes GK rats were divided into three groups, that diabetes mellitus group of 8 weeks(DM8 group),16 weeks(DM16 group),24 weeks(DM24 group),each group 5.The expression of GABAA-R and Glu-R1 in the retinal tissue were detected by Western-blot.GABAA-R and Glu-R1 mRNA expression in the retinal tissue were analyzed by semi-quantitative reverse transcription polymerase chain reaction (RT-PCR) technique. And the localization of GABAA-R and Glu-R1 in the retinal tissue of type 2 diabetic rats were detected by immunohistochemistry.Results1 Western-blotCompared with the control group:the levels of GABA and Glu protein were higher in DM8 group and DM16 group while weaker in DM24 group; the levels of GABAA-R protein were weaker in DM8 group and DM16 group while higher in DM24 group; the levels of Glul-R protein were higher in DM16 group while weaker in DM8 group and DM16 group2 immunohistochemistry.1) GABA the control group and DM rat retinal tissue expression of GABA lied in the cytoplasm of inner nuclear and outer nuclear layer, inner plexiform layer2) GABAA-R rat retinal tissue expression of GABAA-R lied in the cytoplasm of inner nuclear and inner plexiform layer3) Glu rat retinal tissue expression of Glu lied in the cytoplasm of ganglion cells, inner nuclear layer, outer nuclear layer and nerve fiber layer, inner plexiform layer4) Glu-R1 rat retinal tissue expression of Glu-R1 lied in the cytoplasm of ganglion cells, inner nuclear layer, and nerve fiber layer, inner plexiform layer; and it also expressed in outer nuclear layer in the DM8 group and DM16 group3 RT-PCR1) GABA and GABA-R The expression of GABA and GABA-R mRNA were all stronger in DM group than those in control group, the highest level in 16W and the lowest level in 24W2) The expression of Glu-R mRNA were gradual lower in DM group than those in control group3) Glu Compared with the control group, the expression of Glu mRNA were stronger in DM8 group and DM16 group while weaker in DM24 group, the highest level in 8WConclusion1 As excitatory and inhibitory neurotransmitters, GABA and Glu were involved in the invasion and development of retinopathy of Type II dibetes2 The activity of GABAA-R and Glu-R1 related with different phases of DR3 GABA and GABAA-R may delated the progress of DR in the early stage4 Glu-R1 may involved the neurotoxicity induced by Glu in the development of DR... | Keywords/Search Tags: | diabetic retinopathy, neurotransmitters, GABA, GABAA-R, GluR1, Glu, TypeⅡDiabetes Mellitus, pathogenesis | PDF Full Text Request | Related items |
